Purifying selection constrains the evolution of Juquitiba virus in wild Oligoryzomys nigripes communities.
Juquitiba virus (JUQV) is endemic in Oligoryzomys nigripes across several South American countries and causes hantavirus pulmonary syndrome when transmitted to humans via infectious saliva or excreta. We developed a next-generation sequencing (NGS) pipeline to generate the first complete reference genome for assessing the genetic diversity of JUQV in Oligoryzomys populations inhabiting the Mbaracayú Biosphere Reserve within the Atlantic Forest of Paraguay. From 32 additional Oligoryzomys specimens, we obtained 17 S- and M-segment viral RNA (vRNA) genomes from lungs with 94-100% sequence coverage and 101 additional vRNAs with ≥80% genome coverage and ≥500x sequence depth from saliva, urine, lungs, heart, kidney, liver, and spleen. Phylogenetic and phylogeographic analyses showed that the Paraguayan JUQV is genetically distinct from the Brazilian JUQV lineage. Shannon entropy calculations of genetic diversity revealed that saliva and lung samples had higher entropy values than urine, kidney, spleen, and heart samples. The greater genetic diversity was driven in part by greater nucleotide, but not amino acid, diversity in persistently infected rodent samples compared to acutely infected ones. Genetic diversity varied across collection sites, although, given the continuous habitat matrix, there was no apparent reason for these differences. Fixed Effects Likelihood analysis of lung, saliva, and urine sequences suggested that purifying selection was the primary driver of evolution, with no evidence of positive selection. Only three of the 29 codons in the N protein and the glycoprotein (GP) were under purifying selection, and only Gn harbored nonsynonymous mutations. We tested two of the nonsynonymous mutations within the Gn for their effect on entry into Vero cells using VSV-pseudotyped JUQV GP; however, only V504I resulted in a significant reduction in entry compared to wild-type Gn. In summary, tissue source, field locale, and persistent infection were clear drivers of virus evolution.
Detection of Anti-Orthohantavirus Antibodies in Febrile Patients in India.
Orthohantaviruses are a genus of zoonotic viruses from the family Hantaviridae, which is primarily known for their ability to cause severe human diseases. The virus is primarily known for causing two major human diseases, hantavirus pulmonary syndrome and hemorrhagic fever with renal syndrome. However, the frequency of this infection remains largely unknown. Hence, the aim of this study was to diagnose patients infected with Orthohantavirus in India. We tested serum samples from 216 febrile patients, presented at Lisie Hospital, Kochi, for positive anti-orthohantavirus IgM and IgG antibodies. All the participating patients were negative for dengue and scrub typhus IgM antibodies. In total, 16.20% (n = 35) of the febrile patients were positive for anti-orthohantavirus IgM and 11.57% (n = 25) for anti-orthohantavirus IgG antibodies. Anti-orthohantavirus IgM and IgG both were detected in 4.63% (n = 10) of the enrolled subjects. In total, 51.43% of the IgM-positive patients had a final diagnosis of either viral fever or acute febrile disease, with only two patients reported to have contact with rodents, and none of these patients had traveled outside of their place of residence. Orthohantavirus infections may be endemic in India, as their hosts are omnipresent. However, the lack of proper diagnostic tools and limited awareness of these emerging infections amongst doctors in India reduces the diagnosis of the disease. Therefore, there is a pressing need for educating healthcare providers about the circulation of orthohantaviruses in India. There is an urgent need for the development of serotype-specific, affordable, and accurate diagnostic tools for early diagnosis of orthohantavirus infections.
Beliefs and socio-cultural perspectives on hantavirus in a rural community in Panama: An ethnonursing study.
Hantavirus pulmonary syndrome is a zoonotic disease that has been present in the Americas since 1993 and in Panama since the early 2000s. The disease is transmitted to humans through the inhalation of aerosols containing viruses from feces, urine, and saliva from asymptomatic, infected rodents. While various climatic, ecological, social and cultural factors have been associated with hantavirus transmission, research on beliefs and socio-cultural perspectives regarding its care and prevention remains limited. This study addresses this gap by exploring the beliefs, socio-cultural context and care practices related to hantavirus disease in an endemic community. Guided by the One Health approach and Leininger's Theory of Culture Care Diversity and Universality, an ethnonursing approach was used in a hantavirus-endemic community in Panama from September 3, 2021, to August 25, 2022. The study involved 30 participants, including 11 key participants (community members with personal or familial experience of hantavirus) and 19 general participants (community authorities and health professionals). Data were collected through in-depth interviews, participant observation and field notes. Leininger's four phases of ethnonursing analysis were followed using ATLAS.ti. Five themes were identified: (1) hantavirus develops within the context of the community, with its own expressions, meanings and lifestyles; (2) living in and of the land: what we are and what defines us; (3) hantavirus prevention and its challenges: what we do and what we propose; (4) God, the higher power that helps, protects, and gives strength and resilience in daily life and in the face of hantavirus disease; and (5) sharing within the family: joys, longings, celebrations and concerns about hantavirus. Based on the study findings, we propose a model of culture care decision and action modes for hantavirus prevention, emphasizing the importance of implementing relevant, sustainable, and culturally aligned prevention practices in the affected communities.
Virological characterization of a new isolated strain of Andes virus involved in the recent person-to-person transmission outbreak reported in Argentina.
On November 2, 2018, a person-to-person transmission outbreak of Andes virus (Orthohantavirus andesense) began in the small town of Epuyén, Argentina. The strain demonstrated a high capacity for sustained transmission among the human population requiring the implementation of quarantine measures, rigorous contact tracing, isolation of close contacts, and active clinical monitoring to prevent further spread. In this study, we report the isolation of this strain, which we name the ARG-Epuyén strain, directly from a clinical sample after just three passages in cell culture. Complete sequencing revealed only a single amino acid change post-isolation, suggesting that this strain can be considered a non-adapted wild-type Andes virus, marking a critical step toward the development of medical countermeasures against this emerging pathogen. The pathogenicity and transmissibility potential of ARG-Epuyén were evaluated in hamsters, the only animal model for Hantavirus Pulmonary Syndrome. Additionally, this strain was compared with Andes/ARG, an ANDV strain previously isolated from the same geographical area in the Argentinian Patagonia, from a rodent specimen. Our findings revealed high infectiousness and efficient hamster-to-hamster transmission through direct contact experiments, although ARG-Epuyén appeared to be less pathogenic than Andes/ARG.
A systematic review of the distribution and prevalence of viruses detected in the Peromyscus maniculatus species complex (Rodentia: Cricetidae).
The North American Deermouse, Peromyscus maniculatus, is one of the most widespread and abundant mammals on the continent. It is of public health interest as a known host of several viruses that are transmissible to humans and can cause illness, including the acute respiratory disease Hantavirus pulmonary syndrome (HPS). However, recent taxonomic studies indicate that P. maniculatus is a complex of multiple species, raising questions about how to identify and interpret three decades of hantavirus monitoring data. We conducted a systematic review investigating the prevalence and spatial distribution of viral taxa detected in wild populations allocated to P. maniculatus. From the 49 relevant studies published from 2000 to 2022, we extracted and analyzed spatial occurrence data to calculate weighted populational prevalences for hantaviruses. We found that detection efforts have been concentrated in the Western United States and Mexico with a focus on the spread of Sin Nombre virus (Orthohantavirus sinnombreense), the primary causative agent of HPS. There are significant gaps in the existing literature both geographically and regarding the kinds of viruses being sampled. These results are significantly impacted by a recent taxonomic split of P. maniculatus into four species, and we were able to update 94% of hantavirus observations to reflect this change. Investigating the uncertain, and likely multiple, phylogenetic histories of these viral hosts should be a key emphasis of future modeling efforts.
