A hipófise é considerada uma "glândula mestra", pois secreta hormônios que controlam o funcionamento de outras glândulas, sendo grande parte de suas funções reguladas pelo hipotálamo.

Non-invasive host transcriptome and HPV oncogene expression map the molecular landscape of HPV-driven cervical lesions.
Cervical cancer remains a significant global health burden. Current screening methods are not yet capable of detecting molecular alterations preceding cytological abnormalities. In this study, we performed integrative transcriptomic profiling of 132 HPV-positive cervical Pap Smear specimens (NILM, ASCUS, LSIL, HSIL), combining HPV genotyping, and E6/E7 mRNA quantification to map molecular progression. Our analysis revealed stage-specific signatures: NILM displayed a "stealth infection" profile marked by upregulated protein synthesis and growth signaling (EGFR/ERBB2) alongside immune suppression. ASCUS presented a critical tipping point with introduction of early oncogenic drivers (CCND1, SHH), while LSIL prioritized viral productivity with suppressed antimicrobial defenses (MPO, DEFA1). HSIL was distinct from earlier stages and defined by cell cycle hyperactivation (CDK1, PLK1), replication licensing (MCMs), and epithelial dedifferentiation. Pathway crosstalk analysis demonstrated minimal overlap between HSIL and earlier stages (OC < 0.07), highlighting molecular discontinuity during malignant transformation. Additionally, E6/E7 mRNA Ct levels were significantly associated with lesion severity (X2 = 24.407, df = 9, p = 0.003), indicating higher viral mRNA expression are associated with more severe cytological abnormalities. These findings highlight the transformative potential of transcriptomic profiling in cervical cancer prevention, offering stage-specific biomarkers to refine risk stratification. By integrating transcriptomics profiling with current clinical testing, clinicians can distinguish transient infections from high-risks lesions likely to progress. This combined approach addresses the critical limitations of morphology and DNA-based methods, enabling more precise therapeutic interventions and reducing unnecessary overtreatment, and the risk of undertreatment or dismissal of high-risk cases.
Discovering the abnormalities and functional importance of ferroptosis-related molecules in cervical cancer.
Ferroptosis is an iron-dependent nonapoptotic form of cell death that links iron, lipid, and glutathione levels to a variety of disease-related activities. However, the characteristics of ferroptosis in cervical carcinoma (CC) are poorly understood. We acquired raw data on CC cohorts from The Cancer Genome Atlas (TCGA) and the Gene Expression Omnibus (GEO) database. Key genes were identified using differential gene expression analysis and intersected for further immune infiltration, transcription regulation, gene set variation analysis (GSVA), gene set enrichment analysis (GSEA), and drug sensitivity analysis. We also used immunohistochemical (IHC) staining to confirm the expression of important genes in cervical cancer tissue and their prognostic relevance. Finally, gene silencing and cell coculture experiments were used to verify the biological functional mechanism and its role in the tumor microenvironment (TME). Through bioinformatics analysis, we discovered that GCH1 and H1.2 are key ferroptosis-related molecules in cervical cancer. GCH1 and H1.2 could act as useful prognostic markers in cervical cancer, and in addition to their connection with the tumor microenvironment, the possible transcriptional regulatory network, hallmark pathways and chemotherapy sensitivity were also clarified. IHC of the tissue microarray (TMA) and immunofluorescence spatial distance evaluation revealed that GCH1 was more highly expressed in cervical cancer tissue than in paracarcinoma tissue. For patients with cervical cancer, higher GCH1 expression corresponded to a lower M2 cell proportion and a higher M1/M2 ratio as well as a greater GCH1-M2 distance. Silencing GCH1 in SiHa cells blocked the cell cycle, promoted apoptosis, and inhibited the migration and invasion abilities of the cells, possibly through the inhibition of the phosphorylated PI3K/AKT/mTOR pathway. Coculture of the cells with macrophages revealed that the silencing of GCH1 led to decreased expression of tumor necrosis factor (TNF), a biomarker of M1 macrophages. In this study, we performed a thorough investigation of ferroptosis-related genes and identified the functional complexity of GCH1 during tumorigenesis in cervical cancer.
Infertility in an American Quarter Horse mare with cervical duplication.
Cervical duplication is a disorder of sexual development (DSD) that may be present in the absence of chromosomal aberrations. It is a rare cause of infertility in mares and is attributed to the failure of the Müllerian ducts to fuse properly, leading to the persistence of the median walls along the whole length of the cervix. This case report involves a 13-year-old American Quarter Horse mare with cervical duplication. Prior to this diagnostic, the mare was bred multiple times, but pregnancy was not achieved. The mare presented for a breeding soundness evaluation. During the examination an aberrant cervical conformation was palpated and further investigated. Cytogenetic analysis and a PCR for the Y-linked SRY gene and the X-linked androgen receptor gene was performed. A normal 64,XX, SRY-negative karyotype was determined. Infertility was associated with cervical abnormality, affecting in some degree uterine clearance causing accumulation of fluid seen on ultrasound.
Cytological versus histological results in ASCUS cervical dysplasia - a retrospective study.
The cytological category of atypical squamous cells of undetermined significance describes cellular abnormalities that are more severe than inflammatory changes, but are quantitatively or qualitatively insufficient to be included in the squamous intraepithelial lesion category. This study aims to determine the risk level for the presence of high-grade dysplasia in patients with this cytological abnormality. We retrospectively searched our database for women with cytologically proven atypical squamous cells of undetermined significance lesions between January 2020 and June 2024. A total of 104 patients who had undergone colposcopy-directed biopsies were included in the study. Among the 104 women with confirmed atypical squamous cells of undetermined significance cytological lesions who had undergone biopsies, 56 cases (53.8%) were negative, while 48 cases (46.2%) demonstrated cervical intraepithelial neoplasia. Cervical intraepithelial neoplasia 2+ was present in 23.1% (N = 24) of the cases. Furthermore, among 77 human papillomavirus positive women, the precancerous condition was not histologically confirmed in almost half of the cases (N = 36), while in 16 of the 23 human papillomavirus negative biopsies, the precancerous condition was not confirmed. Human papillomavirus status was unknown in 4 cases. We identified a 23.1% presence of cervical intraepithelial neoplasia 2+ lesions in patients with atypical squamous cells of undetermined significance cytological findings. Our study also suggests a lower specificity, but a better negative predictive value of the human papillomavirus test in detecting cervical intraepithelial neoplasia in these patients.
High-risk HPV and bacterial STIs in a primary screening population in rural Hainan, China: prevalence, co-infection, and association with cervical abnormalities.
With the expansion of cervical cancer screening in China, clarifying the epidemiology of high-risk human papillomavirus (HR-HPV) and co-infecting reproductive tract bacterial pathogens in underserved regions is increasingly important. We investigated the prevalence and co-infection patterns of HR-HPV and bacterial pathogens (Chlamydia trachomatis, Ureaplasma urealyticum, Neisseria gonorrhoeae) among rural women in Hainan, and examined their associations with cervical epithelial lesions to inform optimized risk stratification for this population. Within a screening cohort of 8,925 women in Hainan (May-October 2025), we employed a nested case-control design enrolling 869 HR-HPV-positive cases and 473 negative controls, which were randomly selected from HR-HPV-negative women, with age adjustment applied in statistical analyses. Participants underwent genotyping for HR-HPV and bacterial sexually transmitted infections (C. trachomatis, U. urealyticum, N. gonorrhoeae). Cervical abnormalities were assessed via ThinPrep Cytologic Test and subsequent histopathology. HR-HPV infection was associated with older age and lower educational attainment (p < 0.001). Among HR-HPV subtypes, single-type HR-HPV infection predominated (83.08%). U. urealyticum was the most prevalent reproductive tract bacterial pathogen (45.75%) and was more frequently co-detected in HR-HPV-positive women than in controls (p < 0.001). Network analysis further identified U. urealyticum as a central hub, showing strong co-occurrence with HR-HPV52 and HR-HPV58. Clinically, C. trachomatis was independently associated with higher odds of atypical squamous cells of undetermined significance (aOR = 2.82, p = 0.025), whereas U. urealyticum was associated with an increased risk of cervical intraepithelial neoplasia grade 1 (aOR = 1.79, p = 0.040). This study highlights a distinct co-infection ecosystem centered on U. urealyticum in rural women, where C. trachomatis and U. urealyticum drive differentiated risks for early cervical lesions. Consequently, public health strategies should prioritize this underrepresented rural population. Integrating targeted bacterial STIs testing into HPV screening could significantly enhance risk stratification and intervention efficiency in settings with limited resources.
