Observations of the developmental stages of Coquillettidia perturbans (Diptera: Culicidae) with special reference to the pupal trumpets.
Coquillettidia perturbans (Walker) is a North American mosquito species of medical and veterinary importance that has been implicated as a vector of eastern equine encephalitis virus. Larval and pupal C. perturbans possess highly specialized respiratory appendages that enable them to respire from air vesicles in the hollow roots and shoots of emergent aquatic vegetation, which means that the larval and pupal stages have remained largely understudied. Field-collected C. perturbans larvae, pupae, and adults were studied under laboratory conditions in an attempt to establish a laboratory colony. Blood-fed adults successfully laid egg rafts in containers provisioned with water hyacinth plants (Pontederia crassipes). Larvae and pupae were maintained on hydroponically grown wheat seedlings (Triticum sp.). Images and descriptions of egg rafts, larvae, and pupae are provided with special reference to hatch success, larval instar traits and pupal trumpet morphology.
Isolation of Madariaga Virus (MADV) in a Horse Coinfected with Equine Infectious Anemia in Venezuela: A Review of MADV Circulation in the Country.
Madariaga virus (MADV), formerly known as the South American variant of Eastern Equine Encephalitis virus (EEEV), is an alphavirus that belongs to the Togaviridae family and has been periodically infecting equids in Venezuela since its first identification in 1975. This study reports the isolation and molecular characterization of MADV isolated from a horse in December 2024 in the context of MADV cases reported in Venezuela. Antibodies to the rabies virus were detected by indirect immunofluorescence, and to the Equine Infectious Anemia virus (EIAV) by passive immunodiffusion. MADV RNA was detected by qRT-PCR. The sequence of the complete viral genome was obtained by next-generation sequencing. The sequence of this virus was highly similar to that of the only human case of MADV reported in the country in 2016, as well as to a sequence of a virus isolated from a horse in Colombia in 2002. The horse was found to be co-infected with EIAV. The continuous circulation of MADV in Venezuela warrants reinforcing the preventive measures against these alphaviruses, which ignore borders, and may cause important animal and human health concerns.
Investigation of neurotropic arboviruses in wild and domestic animals in Amazon region, 2023-2024.
Arboviruses represent a potential threat to global public health due to their ability to infect various vertebrate hosts and vectors, as well as their adaptability to diverse ecosystems, allowing them to expand geographically across continents. The present study aimed to describe the molecular epidemiology of encephalitic arboviruses, including Venezuelan equine encephalitis virus (VEEV), Eastern equine encephalitis virus (EEEV), Western equine encephalitis virus (WEEV), West Nile virus (WNV), Saint Louis encephalitis virus (SLEV), Toscana virus (TOSV), and Rift Valley fever virus (RVFV) in nervous tissue samples from domestic and wild animals from the Northern region of Brazil, between February 2023 and June 2024. Samples negative for rabies virus were analysed by reverse transcription real-time polymerase chain reaction (RT-qPCR) targeting Alphavirus, Orthoflavivirus and Phlebovirus species.Positive samples were subjected to viral isolation in cell culture and whole-genome sequencing using next-generation sequencing.Phylogenetic and molecular clock analyses were performed to characterise viral lineages and evolutionary relationships. Two samples tested positive for arboviruses by RT-qPCR: one SLEV sample from the state of Amazonas, which showed a low viral load, preventing virus isolation and sequencing, and one Madariaga virus (MADV) sample from the state of Pará, which could be isolated and sequenced. The isolated agent belongs to lineage III of EEEV, showing most similarity to strains from Guyana and Argentina. The present study detected two arboviruses in animals, suggesting its circulation in the study area.
Impact of mutations affecting 4'-fluorouridine susceptibility on fitness and treatment outcomes for Venezuelan equine encephalitis virus.
Venezuelan equine encephalitis virus (VEEV) is a prototypical encephalitic alphavirus. Members of the Alphavirus genus are found across the globe, transmitted by arthropod vectors, and cause significant disease burdens in humans and animals. There are currently no FDA-approved antivirals for human use against any member of the Alphavirus genus. While a vaccine exists against chikungunya virus (CHIKV), a member of the arthritogenic alphaviruses, FDA-approved vaccines are not available for other members of this genus, particularly the encephalitic alphaviruses such as VEEV, Eastern equine encephalitis virus, and Western equine encephalitis virus. 4'-Fluorouridine (4'-FlU, EIDD-2749) was recently identified as a broad-spectrum antiviral against multiple RNA viruses, including alphaviruses. 4'-FlU can potently inhibit VEEV-TC83 replication, with submicromolar potency in cell culture. However, the emergence of antiviral resistance represents a hurdle for antiviral drug development and the implementation of effective treatment strategies. Here, we have identified novel mutations in the VEEV nsP4 RNA-dependent RNA polymerase that reduce susceptibility to 4'-FlU, including P187A, Q191L, L289F, and T296I. We rebuilt each mutation in recombinant VEEV-TC83 and characterized the effects of these mutations on fitness and pathogenicity. In addition, we assessed the impact of mutations reducing sensitivity to 4'-FlU in a mouse model. Although mutations against 4'-FlU arise quickly in vitro, treatment can still alleviate severe disease and lethal encephalitis. Together, these data highlight the promising therapeutic potential of 4'-FlU for the treatment of alphavirus encephalitis.IMPORTANCEVenezuelan equine encephalitis virus (VEEV) is a mosquito-spread virus that can cause encephalitis in people and animals. There are no FDA-approved countermeasures to treat VEEV infections in humans. 4'-Fluorouridine (4'-FlU) is currently being developed to treat multiple viral infections, including VEEV. A major problem with antivirals is the appearance of virus populations that are less susceptible to treatment. In this study, we treated infected mice with 4'-FlU and measured how well the compound inhibited virus replication and prevented severe disease. In addition, we identified mutations in VEEV's polymerase that confer reduced susceptibility to 4'-FlU. We then assessed if viruses encoding for these mutations were still pathogenic. Although VEEV can develop mild resistance to 4'-FlU in vitro, administration of 4'-FlU still reduced severe disease and prevented lethality in the animals infected with viruses that possess mutations that decrease susceptibility to 4'-FlU. These results suggest that 4'-FlU has strong potential as a future treatment for alphavirus infections.
MOSQUITO SPECIES OF NEW YORK STATE: AN UPDATED CHECKLIST WITH PUBLIC AND VETERINARY HEALTH SIGNIFICANCE.
We present an updated checklist of mosquito species in New York State, integrating historical records, modern surveillance, and recent literature. A total of 68 species across 10 genera are documented, including invasive taxa (Aedes albopictus, Ae. japonicus) and historically rare species (Orthopodomyia alba, Or. signifera). This synthesis emphasizes species of public and veterinary significance, such as Culex pipiens and Culiseta melanura, vectors of West Nile and eastern equine encephalitis viruses. Analysis of more than 3 million specimens collected since 1999 confirms the absence of Ae. aegypti, historically introduced but unable to persist in New York. Urbanization, container-breeding ecology, climate change, and global commerce continue to shape mosquito distributions and pathogen risk. This checklist bridges historic and current data, providing a reliable reference for surveillance, vector control, and future research.
