Assessment of a baloxavir marboxil treatment protocol for high pathogenicity avian influenza in Okinawa Rails, an endangered species endemic to Japan.

High pathogenicity avian influenza (HPAI) is a highly infectious and lethal disease of birds that causes systemic symptoms and has been spreading globally, including in Japan. The Okinawa rail (Hypotaenidia okinawae), a flightless bird endemic to Japan, is classified as an endangered species on the Red List. In 2004, the Ministry of the Environment of Japan began implementing a conservation breeding program for Okinawa rails, focusing on maintaining the species' genetic diversity, captive breeding, and reintroduction to the wild. Given the potential for significant losses due to HPAI in Okinawa rails, the establishment of a treatment protocol as a preparedness measure is essential. The aim of this study was to determine an appropriate treatment method for HPAI in Okinawa rails using baloxavir marboxil (BXM), a drug shown to be effective in an avian laboratory model of HPAI virus infection. Single oral administration of BXM at 2.5 or 12.5 mg/kg did not produce plasma concentrations sufficient to achieve the expected therapeutic efficacy. Therefore, oral administration was deemed inadequate for generating the desired pharmacological effects. Consequently, subcutaneous administration of BXM to Okinawa rails at a dose of 2.5 or 7.5 mg/kg was explored as an alternative protocol, which resulted in higher systemic exposure compared with oral administration. Furthermore, plasma concentrations were maintained at therapeutically relevant levels up to 24 hours after subcutaneous administration at 7.5 mg/kg, with mild and reversible injection site irritation the main adverse effect. Based on these results, subcutaneous administration of BXM is proposed as a viable treatment protocol for HPAI in the conservation of endangered Okinawa rails.

Segregate, Test, Observe and Persevere (STOP): strengthening ex situ breeding programs for biodiversity in zoos amid highly pathogenic avian influenza threats - a case approach.

Zoos with avian populations are vulnerable to incursions of the high pathogenicity avian influenza virus (HPAIV) H5Nx due to the free-range husbandry, bird population density and shared open water areas between zoo birds and wild waterfowl. An outbreak of HPAIV H5N1, clade 2.3.4.4b, genotype EA AB, at the Zoological Garden Karlsruhe, Germany, in 2022, was managed applying legal restriction measures but exceptionally exempting culling orders for RT-qPCR-positive but clinically healthy birds. A critical factor in the zoo's response was the implementation of a segregation concept approved in advance. The entire bird population could be rapidly separated into epidemiological housing units (epUs) cared for by separate staff. A total of 79 birds initially tested RT-qPCR-positive, but only 21 (26.6%) clinically diseased birds had to be euthanized or succumbed. Seroconversion amounted to 94.8% of the remaining 58 birds. Extensive RT-qPCR investigations of 3,634 samples confirmed infections remained confined to three initially infected epUs out of a total of 25. Spread of virus in the infected epUs was limited to 3 weeks after segregation. In the cohort of infected pelicans, surviving individuals remained seropositive with elevated levels of H5-specific antibody titers for the following 2 years suggesting ongoing protection. The described science-based control measures rested on a legally binding yet interpretive statement regarding the pertinent animal health legislation are exemplary for managing an outbreak of high pathogenicity avian influenza (HPAI) in zoos.

Evaluation of the inactivation of avian influenza virus in duck meat through electron beam irradiation.

The global spread of highly pathogenic avian influenza (HPAI) among poultry, wild birds, and mammals has increased the risk of infection in felines, including domestic and stray cats. Wild felids are particularly susceptible to H5N1 HPAI virus infection because they may consume the carcasses of infected birds. Fatalities associated with the consumption of raw pet foods prepared from poultry meat have been reported in both the United States and Korea. Given that high titers of avian influenza (AI) viruses have been detected in poultry meat, effective sterilization methods are essential to ensure the safety of raw pet food products. In this study, we evaluated the efficacy of electron beam (E-beam) irradiation-an established method for inactivating hazardous microorganisms in raw pet food-against duck meat experimentally contaminated with avian influenza (AI) virus. Shredded duck breast meat was inoculated with the low-pathogenic H5 subtype avian influenza virus A/Geese/Korea/H277/2022 (H5N3) and subjected to E-beam irradiation at doses ranging from 0 to 40 kGy. The D10 values calculated based on the nominal irradiation dose and the actual absorbed dose were 2.58 and 2.34, respectively. Based on the absorbed-dose D10 value, duck meat containing a high viral load (approximately 107.6 EID₅₀/g in the mock-irradiated control) was estimated to require approximately 17.63 kGy for complete viral inactivation.

A Novel PLGA-based nano-adjuvant augments mucosal and systemic immunity in chicks through intestinal targeting and IgA production.

The ongoing dissemination of the H9N2 avian influenza virus presents a significant challenge to the effective control and prevention of avian diseases. Consequently, this study developed a multipleimmune-regulatory adjuvant, PEI-LSP-RA-PLGA, to enhance the immune efficacy of H9N2 vaccines. The adjuvant was prepared by a double-layer nanoparticle preparation technology, with Lagenaria siceraria (Molina) Standl. Polysaccharide (LSP) and retinoic acid (RA) encapsulated into PLGA nanoparticles modified by polyethylenimine (PEI), forming PEI-LSP-RA-PLGA. The nanoparticle size is 200 nm, and the zeta potential is 13 mV. It has good stability and an antigen-sustained-release effect lasting 21 days. PEI-LSP-RA-PLGA was used as an inactivated H9N2 vaccine to immunize chicks. The results showed that PEI-LSP-RA-PLGA nanoparticles could promote the secretion of serum IgG antibodies, It increased by 132.83 % compared with the control group. Intestinal IgA antibodies, it increased by 115.12 % compared with the control group, and various cytokines, enhance the function of immune organs, promote the differentiation of spleen T lymphocytes, and improve the structural morphology of the small intestine. The in vivo imaging findings indicated that the PEI-LSP-RA-PLGA nanoadjuvant exhibited an excellent sustained-release effect at the injection site and demonstrated long-term intestinal targeting. This led to an increase in the number of intestinal IgA+ cells, thus boosting the intestinal immune function. The sequencing and confirmatory test results showed that PEI-LSP-RA-PLGA produced an effective intestinal targeting effect through the CCR9 and CCR6 signaling pathways under the action of the chemokines CCL20 and CCL25. After targeting the intestinal tract, an effective mucosal immune response was generated through the Toll-like receptor pathway, the NOD-like receptor pathway, and the immune network of IgA production. To sum up, PEI-LSP-RA-PLGA is a multi-targeted vaccine adjuvant that can simultaneously induce long-term systemic immunity and intestinal mucosal immunity, showing great application potential in improving the effectiveness of vaccines.

A modelling exploration of potential spatiotemporal risk of high pathogenicity avian influenza virus introduction to Danish dairy herds through the contaminated environment.

Circulation of high pathogenicity avian influenza virus (HPAIV) among cattle in the US as a result from spillover events from wild birds has been reported since 2024. The consequences have reduced animal welfare and milk production and increased human exposure to infections. With HPAIV detected in British sheep in March 2025 and bovine antibodies detected in Dutch dairy cow in January 2026, the threat of a wider spread in European ruminants has been underscored. To timely detect and react against the spread of HPAIV in cattle appearing in new regions, it is essential to assess the spatiotemporal risk from wild birds. We here utilised a previously developed spatiotemporal simulation model for avian influenza in birds in Denmark to assess the weekly risk of introduction to dairy herds in 10 by 10 km squares. We estimated that the peak risk period for potential transmission to dairy herds is between January and March. Areas with the highest risk estimates include northern Jutland and southern Zealand, associated with a dense dairy population and the wild bird migration corridor, respectively. While these results likely represent a worst-case scenario, as many Danish herds are housed indoors during the peak risk months, the model provides a crucial benchmark for environmental infection pressure. The results can be used as a guideline for where and when to monitor for HPAIV introductions to dairy herds in Denmark.
