DMEM and EMEM as alternate growth media for pathogenic Leptospira.
Pathogenic Leptospira species can survive and thrive in a wide range of environments. Distinct environments expose the bacteria to different temperatures, osmolarities, and amounts and sources of nutrition. However, leptospires are mostly cultured in a laboratory setting under in vitro conditions that do not reflect natural environments. This constraint on laboratory cultures limits the applicability of in vitro studies to the understanding of even simple pathogenic processes. Here we report, investigate, and identify a medium and conditions that mimic the host environment during leptospirosis infection, expanding the available in vitro tools to evaluate leptospiral pathogenesis. We quantified genome-wide transcription of pathogenic Leptospira interrogans cultured in different in vitro media compositions and conditions-EMJH at 29 °C and DMEM, EMEM, and HAN at 37 °C and 5% CO2. Using EMJH as standard, we compared gene expression in these compositions to genome-wide transcription gathered in a host environment: whole blood (WB) of hamsters after infection with pathogenic leptospires. Leptospires cultured in DMEM and EMEM media shared 40% and 47% of all differentially expressed genes (DEGs) of leptospires present within WB (FDR < 0.01), while leptospires cultured in HAN media only shared 20% of DEGs with those from WB. Furthermore, gene and pathway expression of leptospires cultured on DMEM and EMEM media exhibited a better correlation with leptospires grown in WB, including promoting expression of a similar leptospiral lipid A profile to the one identified directly in host tissues. Taken together, these results indicate that commercial cell-culture media EMEM or DMEM are better surrogates for in vivo pathogenic studies than EMJH or HAN media in Leptospira. These alternative culture conditions, using media that are a standard supply worldwide, provide a reproducible and cost-effective approach that can accelerate research investigation and reduce the number of animal infections necessary for basic research of leptospirosis.
Functional Validation of the Proteome-Identified LIC_13056 Putative Lipoprotein of Leptospira interrogans and the Potential Role in Pathogenesis.
Leptospirosis is a widespread zoonosis of human and veterinary concern. The etiological agent of the disease is the pathogenic bacteria of the genus Leptospira. Transmission typically occurs through mucosal contact and/or injured skin with the urine of infected animals or contaminated environmental sources. Understanding the biology and pathogenesis of leptospires is the main focus of our study. In this work, we characterized a novel protein encoded by the LIC_13056 gene from L. interrogans serovar Copenhageni, having an OmpA-like domain. We show that this coding sequence (CDS), previously assigned as a hypothetical protein with an unknown function, is capable of binding to the cellular receptor α8 integrin subunit, potentially contributing to kidney colonization. Additionally, the protein bound to both purified and normal human serum (NHS) plasminogen (PLG). In both conditions, PLG bound to protein was able to generate plasmin (PLA). Furthermore, rLIC_13056 interacted with the complement system components C4b, C4BP, C8 and C9. The interaction of recombinant protein to the C9 had a negative impact on C9 polymerization. Taken together, the protein LIC_13056, having an OmpA-like domain, appears to be involved in leptospiral pathogenesis via different stages of the infection process; PLA generation together with the inhibition of the membrane attack complex (MAC) may contribute to the immune evasion mechanism of Leptospira, thus facilitating the infection.
First report of pathogenic Leptospira in Sri Lankan bats: A potential reservoir risk?
The genus Leptospira, classified under the phylum Spirochaetes, includes saprophytic, intermediate, and pathogenic species. Pathogenic Leptospira spp. are the causative agents of leptospirosis, a widespread and often neglected zoonotic disease that causes severe illness in humans, particularly in tropical and subtropical regions. In Sri Lanka, leptospirosis causes annual outbreaks, especially during the monsoon seasons. While rodents are recognized as primary reservoirs, bats have also been identified as potential reservoir hosts. This study aimed to detect and characterize Leptospira species in bats roosting in Wavulgalge cave, Sri Lanka. Urine samples (n = 148) were collected during natural urination from four bat species: Miniopterus fuliginosus (n = 117), Hipposideros speoris (n = 8), Rousettus leschenaultii (n = 10), and Rhinolophus rouxii (n = 13). DNA was extracted and screened for Leptospira using real-time PCR targeting the lipL32 gene. Sixteen samples tested positive, including 14 from M. fuliginosus, one from H. speoris, and one from R. leschenaultii. Positive samples were further analyzed by amplifying additional loci (flaB, secY, and rrs2) for Sanger sequencing and phylogenetic analysis. Sequences were obtained from 12 samples. Leptospira borgpetersenii was identified in M. fuliginosus, while the sequence from R. leschenaultii represented a genetically distinct Leptospira species. Phylogenetic analysis showed clustering with sequences from the same bat host genera reported in other countries, and one L. borgpetersenii sequence clustered with strains previously detected in Sri Lankan hosts, including humans. This study provides the first molecular evidence of Leptospira in Sri Lankan bats and highlights their potential role as reservoirs contributing to the environmental circulation of pathogenic Leptospira.
Bridging the gap: Multi-sector perspectives on human, domestic animal, and wildlife leptospirosis in Ontario, Canada.
Although leptospirosis is one of the most common zoonotic diseases worldwide, limited surveillance and poor coordination between human and animal health sectors have resulted in scarce and disparate data on its occurrence. Strengthening integrated surveillance requires cross-sector collaboration, beginning with the engagement of key organizations. The aims of this study were to 1) determine key health experts' awareness and risk perceptions of leptospirosis and of zoonotic disease surveillance in Ontario, Canada, and 2) examine key components of engagement, such as perceived value and interest, during the initial stages of developing an integrated leptospirosis surveillance framework. A web-based survey was sent to 543 experts in human, animal, and environmental health in Ontario, and analyzed using a mixed-methods approach to identify key factors influencing perceptions of leptospirosis, including views on Leptospira distribution, the impact of human behavior, and the influence of environmental conditions. Leptospirosis was recognized as a health threat in Ontario by 90% (74/82) of respondents, and 91% (70/77) indicated that current surveillance efforts are inadequate. A higher proportion of animal health sector respondents identified leptospirosis as a threat to human (93%, 37/40) and animal health (90%, 44/49) compared to public health sector respondents (76%, 25/33 and 83%, 25/30, respectively). All participants (81/81) acknowledged the benefits of integrated surveillance over the current siloed approach. Our findings highlight that key public and animal health experts perceive leptospirosis as a health threat in Ontario and support more integrated disease surveillance to better respond to this emerging zoonotic pathogen.
Pathogenic Leptospira species identified in dogs and cats during neutering in Thailand.
Pathogenic species of the genus Leptospira cause an underdiagnosed zoonosis in humans and animals called leptospirosis. Animal reservoirs often remain asymptomatic yet shed the active spirochete in urine, making the control of leptospirosis transmission to humans more challenging. Asymptomatic leptospirosis in human companions, such as dogs and cats, resulting in unrecognised infections, has been demonstrated in a few countries. Crucially, the current lack of molecular epidemiology data on Leptospira among companion animals in Thailand underscores the urgent need to investigate transmission dynamics for effective regional control. We investigated the prevalence of Leptospira infection in cats and dogs during neutering in seven provinces across Thailand. The urine samples were screened for Leptospira DNA by PCR targeting the rrs gene and further speciation using the Sanger Sequencing Analysis. The 56/567 (9.9%) animals were positive for Leptospira in the Pathogen clade, including 34/303 (11.2%) dogs and 22/264 (8.3%) cats. The partial rrs gene analysis identified L. interrogans, L. weilii, and L. borgpetersenii (4.4%) as well as Pathogen subclade 2 species (1.4%). Notably, this study reports the first molecular detection of L. yasudae (1.0%) in companion animals in Thailand. The identification of these three key pathogenic Leptospira species, common causes of human leptospirosis in Southeast Asia, in clinically healthy owned and free-roaming dogs and cats, suggests the risk of human leptospirosis in the areas investigated. These companion animals, often living in close contact with human, may contribute to daily risks. Therefore, enhanced surveillance and vaccination programs for dogs and cats, coupled with targeted public awareness campaigns, are critical for mitigating the risk of human infections.
