Automated identification of bones and joints in whole-body CT-scan images of pigs.
Computed tomography (CT) images provide fast and accurate non-invasive measurements of anatomy, which are crucial in pig breeding. In the commercial breeding program of Topigs Norsvin, the CT-images are mainly used for estimation of carcass value and for scoring the severity of osteochondrosis lesions in joints. This study presents the first major step towards automated detection of osteochondrosis, using CT images. This involves an anatomic segmentation model that can segment 29 classes of different tissues, like individual bones, muscles, and organs. The algorithm then identifies major joints in the fore- and hindlimbs by detecting the center points of the joints, and this method is validated against manually labeled data. Average distance between labeled and predicted center points was 29 mm (SD = 13mm). The next step will utilize these center points to create bounding boxes for local segmentation models to focus on relevant subsets of voxels, enhancing the detection of lesions in joints. This approach aims to improve the efficiency and accuracy of osteochondrosis quantification in CT scan images of pigs, ultimately -benefiting pig breeding programs by providing detailed and automated phenotypes. We have developed a new computer-based method to automatically identify bones and joints in CT scans of pigs. These CT scans are used in pig breeding to measure body composition and detect joint diseases like osteochondrosis, a condition that can cause pain and leg problems in pigs. Until now, we have manually inspected CT images to find and measure the severity of joint lesions. This was slow and expensive. The new method uses artificial intelligence (AI) to automatically recognize 29 different tissues, including bones, muscles, and organs. It then pinpoints the exact location of major joints like shoulders, elbows, knees, and ankles. Tests show that the joint detection is very close to expert labeled data. By focusing on these joints, the system can help future AI models measure joint lesions more accurately and efficiently. This breakthrough could make pig breeding program more sustainable and cost-effective, by helping breeders select animals with better joint health, using smart technology instead of manual scoring.
Therapeutic effects of platelet-rich plasma on cartilage in experimental swine models of leg weakness.
This study examined the effect of platelet-rich plasma (PRP) administration on the repair of articular cartilage damage in domestic pigs with experimentally induced leg weakness. The left hind stifle joints of six 56-day-old castrated domestic pigs were artificially injured; PRP was injected into the stifle joints of three pigs. Histopathological analysis of dissected articular cartilage revealed that all specimens were lightly stained with Alcian blue, and regions containing immature chondrocytes or chondrocyte clusters indicated cartilage repair. The average cartilage thickness was 189.44 μm in the control group and 386.53 μm in the treatment group, showing a significant difference (P≤0.028). These findings suggest that PRP administration promotes articular cartilage repair in domestic pigs.
From growing pains to growing evidence: a 2025 update on novel insights in lower limb osteochondroses.
Osteochondrosis encompasses a heterogenous group of pathologies affecting endochondral ossification in the growing child and adolescent. The cause of each disease is multifactorial, though many are often related to overuse injury, and may be epiphyseal, physeal, or apophyseal. Identification and treatment of this group of disorders is complex, thus this review aims to briefly describe common pathologies, their management, and highlight novel developments within the field. Machine learning as well as advanced diagnostic tools for more precise evaluation and prognostication of osteochondroses have been studied including perfusion MRI in Legg-Calvé-Perthes disease. Novel treatments include leukocyte-rich platelet-rich plasma (LR-PRP), which offer promising improvements in pain and function in Osgood-Schlatter disease. Surgical technique studies have begun to examine optimal operative management of Freiberg's disease. The osteochondroses are an often-self-limiting spectrum of pathologies affecting the physis in children and adolescents that may be managed with conservative treatment, though some require surgical intervention. Advances in imaging, prognostication tools, and treatment modalities support earlier and accurate diagnoses, as well as better informed treatment decisions.
Automated Joint Space Width Assessment in Patients Treated for Juvenile Osteochondritis Dissecans of the Distal Femur: A Cross-Sectional Study and Systematic Review of the Literature.
Background/Objectives: Juvenile osteochondritis dissecans (JOCD) of the knee is commonly treated using conservative or joint-preserving surgical techniques. While clinical outcomes are generally favorable, the risk of early cartilage degeneration remains unclear. Joint space width (JSW) on weight-bearing radiographs serves as an indirect marker of cartilage health. Artificial intelligence (AI)-based JSW assessment may enable sensitive and reproducible detection of early degenerative changes. Methods: This cross-sectional feasibility study included 21 skeletally immature patients treated for JOCD of the distal femur between 2002 and 2017. Treatment modalities comprised conservative management, retrograde drilling, and fragment refixation. Fully automated JSW measurements were performed on standardized anteroposterior knee radiographs using a validated AI-based software IB Lab KOALA™, Version 2.4. JSW of the affected compartment was compared with the contralateral knee and between treatment groups. Clinical outcomes were assessed using the Lysholm Knee Scoring Scale and the International Knee Documentation Committee (IKDC) score. Additionally, a systematic review of the literature on post-treatment degenerative changes following OCD therapy was conducted according to PRISMA guidelines. Results: Compared with manually reviewing images, the software IB Lab KOALA™, Version 2.4 as easy to implement. AI-based analysis revealed no significant differences in JSW between the affected and contralateral knees, nor between treatment modalities. Average JSW exceeded 6 mm in all groups after a median follow-up of 64 (min. 27, max. 177) months. Clinical scores were high and comparable across treatments. A moderate positive correlation was observed between the JSW and Lysholm score, while increasing age and longer follow-up were associated with a reduced JSW. The systematic review identified ten relevant studies, reporting generally favorable long-term clinical outcomes with a low but present risk of osteoarthritis progression. Conclusions: Our AI-based analysis showed no differences in JSW between conservative and joint-preserving surgical treatments of JOCD in the follow-up. This technology can provide a valuable tool for standardized and sensitive radiographic monitoring in young patients.
Dysregulated Lipid and Protein Networks in Osteochondrosis.
Osteochondrosis is an orthopedic developmental disease that affects subchondral bone and articular cartilage in humans and domestic animals, resulting in lameness and joint effusion. Here, we compared the molecular compositions of cartilage from healthy and osteochondrotic equine metacarpophalangeal joints to investigate the pathogenic processes of the disease. This comparison was carried out using an SP3-label-free proteomics workflow, analyzing the differences in protein abundance. Furthermore, mass spectrometry imaging (MSI) was employed for spatially resolved lipid analysis, providing a deeper understanding of osteochondrosis pathogenesis through molecular pathway analysis. Lipid readouts showed increased phosphatidylcholine (PC) lipid species and a downregulation of sphingomyelin (SM) lipids in osteochondrosis biopsies compared to healthy cartilage. Furthermore, spatial analysis of lipids revealed a higher presence of PC lipids in the superficial layer of the cartilage compared with the deep layer. Osteochondrosis-downregulated proteins were mainly involved in extracellular matrix (ECM) organization, protein folding, and hydroxylation, highlighting the importance of ECM imbalance in the osteochondrosis development. Protein-metabolite integration analysis reported a downregulation of glycolysis in the osteochondrosis group, which might lead to chondrocyte hypertrophy and ECM degradation. Our work provides novel insights into the underlying mechanisms associated with the development of osteochondrosis.
