Red panda intestinal organoids for the functional analysis of Lactobacillus salivarius.
Organoids are excellent and reproducible in vitro models for studying physiological mechanisms and modeling diseases in endangered species. However, intestinal organoids of red pandas have not yet been established. This study reports a culture system of intestinal stem cell-derived organoids of red panda that can be passaged, cryopreserved, and resuscitated in vitro. The single-cell RNA sequencing and RNA sequencing analysis showed that red panda intestinal organoids retain tissue-specific cell types and functions. In order to verify the application potential of Lactobacillus salivarius P103 as a probiotic in red pandas, these organoids were used to establish a co-culture system with L. salivarius P103. The results showed that L. salivarius P103 could promote mucin secretion, reduce intestinal inflammation, and inhibit infection induced by Klebsiella pneumoniae 28. The establishment of red panda intestinal organoids provides a long-term, reproducible model system for studying host-microbe interaction mechanisms and decreasing dependence on animal models in wildlife research.

A Reproducible Python-Based Computational Pipeline for Real-Time Ingestion, Advanced Analysis, and Dynamic Reporting of Public Health Data: A Systems Validation Study.
Background The analysis of large-scale public health data is crucial for evidence-based policymaking, but conventional workflows involving manual data handling and static reporting are inefficient and lack reproducibility. There is a need for automated tools that bridge the gap from live data sources to sophisticated, dynamically generated insights. Objective To design, implement, and validate a fully automated Python (Python Software Foundation, Wilmington, DE, USA) pipeline for real-time application programming interface (API)-based ingestion of existing datasets, analysis, and dynamic report generation in public health informatics. Methods We developed a lightweight, Python-only, Word-report-oriented pipeline using packages including Pandas, scikit-learn, statsmodels, and python-docx. The pipeline ingests data from public APIs with automated retries, performs preprocessing, calculates composite health scores, applies K-means clustering (k = 3) for state stratification, and performs correlation analysis. A custom rule-based engine generates dynamic textual interpretations based on statistical results. The final output is a programmatically constructed Microsoft Word (Microsoft Corporation, Redmond, WA, USA) document containing narrative, tables, and embedded figures. The pipeline was tested using India's Health and Family Welfare Statistics 2015 dataset via the data.gov.in API. Results The pipeline executed successfully in approximately 95 seconds, ingesting 37 records. It generated a composite health score, identifying Meghalaya as the top performer (score: 100.0). K-means clustering stratified states into three distinct performance tiers. Correlation analysis revealed a significant negative association between sub-health centre (SHC) infrastructure and specialist availability (r = -0.446, p = 0.01), as well as between 24×7 service availability and auxiliary nurse midwife (ANM) staffing (r = -0.358, p = 0.05), highlighting a systemic disconnect between capital investment in facilities and human resource allocation. A complete Word report including these findings, figures, and tables was automatically generated. Conclusion This automated framework provides a robust, efficient, and reproducible solution for transforming raw public health data into actionable insights and can significantly accelerate data-driven discovery and reporting in public health and bioinformatics. This study validates a computational framework for automated public health data analysis and reporting.
Species Identification and Spatial Diversity Patterns of the Giant Panda National Park (GPNP) in Chengdu, Sichuan, China.
Elucidating the spatial patterns of biodiversity and their driving mechanisms is crucial for predicting the impacts of environmental changes and informing conservation and management efforts. This study investigated the spatial distribution patterns of species in the Chengdu section of the Giant Panda National Park (GPNP), located in the western part of Sichuan Province, China, which provides a wide range of suitable habitats for diverse species and biotic communities. Species trace points data derived from the 4th National Survey on Giant Pandas were used to analyze the diversity and composition of mammals and gallinaceous birds among the five regions. The results indicated that the central regions comprising Dujiangyan Shi (DJYS), Chongzhou Shi (CZS), and Dayi Xian (DYX) exhibited higher α-diversity, whereas Qionglai Shi (QLS) and Pengzhou Shi (PZS) presented lower values across most indices, a pattern that was consistent with expectations of the mid-domain effect. However, Pielou's evenness index showed no significant differences among regions. β-diversity analysis revealed that species turnover was the dominant factor contributing to faunal heterogeneity among regions, with nestedness playing a relatively minor role. These findings highlight the necessity for coordinated conservation among the five regions within the Chengdu section of the GPNP, with a particular focus on improving habitat connectivity and establishing species dispersal corridors to maintain biodiversity. The study provides valuable insights for the development of targeted conservation strategies and the establishment of a long-term biodiversity monitoring system, which will enhance the ecological sustainability of the GPNP and contribute to the protection of isolated giant panda populations and their sympatric species.
How Do Host Plants Mediate the Development and Reproduction of Phytoseiulus persimilis (Acari: Phytoseiidae) When Fed on Tetranychus evansi or Tetranychus urticae Koch (Acari: Tetranychidae)?
In this study, P. persimilis was provided with T. evansi and T. urticae that had been reared on either bean or potato plants to investigate the effects of both prey and host plant species on the predator's growth, development, and fitness. The results indicate that the reproductive potential of P. persimilis populations fed T. evansi was significantly lower than that of populations fed T. urticae from the same host plant (p < 0.01). Phytoseiulus persimilis fed T. evansi that had been reared on potatoes showed poorer performance in oviposition period, post-oviposition period, daily egg production, and total egg production compared to those fed T. evansi reared on beans (p < 0.01). The intrinsic rate of increase (rm) of P. persimilis fed on T. evansi reared on potato was 0.08, which was 55.56% lower than that of populations fed on T. evansi reared on beans. This study sheds light on the complex interactions among host plants, pests, and their natural enemies, thereby providing a theoretical basis for developing more effective and sustainable management strategies against T. evansi that take these intricate ecological relationships into account.
