EnP1 exploits H2Aub-dependent epigenetic reprogramming to promote microsporidia proliferation in host cells.
Microsporidia, as opportunistic parasitic pathogens, constitute a formidable threat to human health. Although the regulatory circuitry of the nucleus-targeted effector EnP1 remains highly intricate and only partially characterized, our study identifies histone H2A as a novel binding partner of EnP1. Furthermore, we demonstrate that both EnP1 overexpression and microsporidia infection induce monoubiquitination of H2A (H2Aub) through downregulation of BAP1 expression. Subsequent mechanistic analyses revealed that elevated H2Aub levels positively correlate with enhanced microsporidian proliferation, whereas attenuation of H2Aub markedly suppresses pathogen expansion. Furthermore, EnP1 orchestrates the enrichment of H2Aub at the SLC7A11 promoter, driving its transcriptional upregulation. Collectively, these findings underscore that EnP1 modulates the ferroptosis state of host cells through H2Aub-mediated epigenetic reprogramming, ultimately facilitating pathogen propagation. This study endeavors to elucidate the critical survival strategies of microsporidia within host cells mediated by EnP1 and to unravel the multifaceted interplay between these pathogens and their hosts.
Detection of Enterocytozoon hepatopenaei in pond-cultured Litopenaeus vannamei shrimp: Unveiling the epidemiological significance of some invasive macrofauna, synergistic diagnostic approach and treatment trial.
Hepatopancreatic microsporidiosis (HPM) is protozoal disease caused by Enterocytozoon hepatopenaei (EHP), causing remarkable economic losses in the shrimp farming due to associated mortalities as well as size variations. The current study aimed to investigate the roles of some invasive macrofauna in the epidemiology of EHP, analyze the associated hepatopancreatic pathologies and check the efficacy of a novel field treatment for the EHP affected pond. A total of 100 L. vannamei were randomly collected from the affected pond before and after treatment trial. Samples were subjected to thorough clinical, parasitological and histopathological examination as well as molecular typing. A total of 5 moon jellyfish and 5 blue swimmer crabs were also collected from the affected ponds and subjected to parasitological examination. Significant size variations among sampled shrimp ranging from 2.1 g up to 20 g together with increased numbers of stunted shrimps were recorded. Behavioral changes such as poor feed response with/without white fecal threads were observed. The EHP spores in hepatopancreatic smears & impressions were identified using conventional methods by Giemsa and modified trichrome staining. The identity of the EHP spores was confirmed by the conventional PCR and sequencing of the PCR product. Histopathology revealed different degrees of degeneration at level of tubular epithelium and inter-tubular tissue together with random appearance of EHP spores. The moon jellyfish and blue swimmer crabs could play a role in EHP infection. The effectiveness of the used immunostimulatory & disinfection treatment protocol was confirmed by the remarkable reduction of the spore numbers in hepatopancreatic tissues.
An Investigation of Microsporidia Species in Humans Living in Düzce and Bolu Provinces of Türkiye.
The literature about microsporidia clearly shows that the prevalence of microsporidia varies at different rates depending on the sampling status and region in the studies. However, the majority of studies have been conducted in immunocompromised individuals. It is vital to determine the prevalence of pathogens in society in order to protect immunocompromised individuals from opportunistic pathogens. Consequently, the present study investigated stool samples from Düzce and Bolu provinces for microsporidia, irrespective of disease group. A study of 400 stool samples from Düzce and Bolu was performed. The samples were first stained with trichrome and examined under a microscope at 100x magnification. DNA was then isolated, amplified with polymerase chain reaction primers, and the samples were identified. The relationship between microsporidia presence and age, sex, and diarrheal status was statistically investigated. Following microscopic examination, microsporidia spores were detected in 25 of the 400 (6.25%). Microsporidia spores were detected in 6.6% (14/212) of the Düzce samples and 5.8% (11/188) of the Bolu samples. Consequently, molecular analysis revealed the presence of microsporidia in 49 of the 400 (12.25%). The positivity rates were found to be 11.3% (24/212) for Düzce and 13.3% (25/188) for Bolu. The spores detected in this study were identified as belonging to the species Enterocytozoon bieneusi. A statistical analysis was conducted, revealing no significant association between the presence of the pathogen and diarrhea complaints, gender, or age. It is seen that the presence of microsporidia in our study is at a level similar to rates reported in immunocompetent individuals around the world. Additionally, a correlation between the presence of microsporidia and complaints has been reported, especially in immunocompromised individuals, but no relationship has been determined with complaints in this study. Furthermore, the present study demonstrated that neither age nor gender exhibited any correlation with the presence of microsporidia. Literatüre bakıldığında, fırsatçı patojen mikrosporidiaların yaygınlığının çalışmalardaki örneklem durumuna ve bölgelere göre farklı oranlarda raporlandığı görülmektedir. Bununla birlikte çalışmaların büyük bir çoğunluğu bağışıklığı yetersiz kişilerde gerçekleştirilmiştir. Bağışıklığı yetersiz kişilerin fırsatçı patojenlerden korunması için toplumda o patojenin yaygınlığının belirlenmesinin çok önemli olduğunu düşünmekteyiz. Bu sebeple, mevcut çalışmada hastalık grubu gözetmeksizin Düzce ve Bolu illerinden gaita numuneleri mikrosporidia açısından araştırılmıştır. Bu çalışmada Düzce ve Bolu illerinde yaşayan insanlardan toplam 400 gaita numunesi temin edilmiştir. Örnekler ilk olarak trikrom boyama ile boyanarak mikroskopta 100x büyütmede incelenmiştir. Yine her örnekten DNA izolasyonu yapılmış ve sonrasında türlere özgü primeler kullanılarak polimeraz zincir reaksiyonu ile tür tespiti gerçekleştirilmiştir. Mikrosporidia varlığının yaş, cinsiyet ve ishal durumu ile ilişkisi istatistiksel olarak araştırılmıştır. Mikroskobik incelemeler sonucunda 400 örneğin 25’inde (6,25) mikrosporidia sporları saptanmıştır. İllerin kendi içerisinde değerlendirilmesi sonucunda Düzce örneklerinin %6,6’sında (14/212) Bolu örneklerinin %5,8’inde (11/188) mikrosporidia sporları görülmüştür. Moleküler analizin sonucunda 400 örnekten 49 tanesinde (12,25) mikrosporidia saptanmıştır. Pozitiflik oranlarının Düzce için %11,3 (24/212) ve Bolu için %13,3 (25/188) olduğu görülmüştür. Tespit edilen sporların Enterocytozoon bieneusi türüne ait olduğu saptanmıştır. Yapılan istatistiksel analiz sonucunda patojenin varlığı ile ishal şikayeti, cinsiyet ve yaşın arasında anlamlı bir ilişki olmadığı görülmüştür. Çalışmamızdaki mikrosporidia varlığının bağışıklığı yeterli kişilerde raporlanan oranlarla benzer düzeyde olduğu görülmektedir. Bu durum, mikrosporidia türlerine maruz kalmanın yaygın olduğunu ve sağlıklı bireylerde herhangi klinik belirti göstermeden enfeksiyonun toplumda sürekli devam ettiğini göstermiştir. Literatürde mikrosporidia varlığı ile gastrointestinal şikayetlerin ilişkili olduğunu bildiren çalışmalar varken tam tersi durumun da varlığı raporlanmıştır. Özellikle bağışıklık sistemi açısından kronik bir rahatsızlığı olmayan kişilerde mikrosporidia türleri kendini sınırlayan ve klinik belirti göstermeyen enfeksiyonlar oluşturması sebebiyle bazı çalışmalarda şikayetlerle mikrosporidia ilişkisinin olmadığının raporlanması normal bir durumdur.
Susceptibility of shrimp Penaeus stylirostris to Ecytonucleospora hepatopenaei and horizontal transmission between P. stylirostris and P. vannamei.
The microsporidium Ecytonucleospora hepatopenaei (EHP) continues to disrupt farmed shrimp production globally by causing growth retardation, chronic mortality, and enhancing susceptibility to other diseases. While the susceptibility of black tiger shrimp Penaeus monodon and Pacific white shrimp P. vannamei to EHP is well known, the susceptibility of Pacific blue shrimp P. stylirostris to EHP has not been demonstrated. To determine the susceptibility of P. stylirostris to EHP, infectious inoculum was directly injected into the hepatopancreas of specific-pathogen-free (SPF) P. stylirostris. At 17 d post-injection, the developmental stages of the parasite were observed in the EHP-injected P. stylirostris, and EHP was detected by real-time PCR. The EHP-injected P. stylirostris were then divided into 2 groups. In the first group, P. stylirostris (n = 9) were cohabitated with SPF P. vannamei (n = 55), and in the second group, the hepatopancreas was excised from EHP-injected P. stylirostris, homogenized, and fed to SPF P. vannamei (n = 12). Both experimental challenge routes led to the horizontal transmission of EHP from P. stylirostris to P. vannamei, but cohabitation resulted in a stronger infection. In a follow-up study, one group of SPF P. stylirostris was fed EHP-infected P. vannamei tissue, and another group cohabitated with EHP-infected P. vannamei. Both groups of P. stylirostris developed EHP infections. These results clearly provide evidence of both P. stylirostris susceptibility to EHP and the transmission potential of EHP between P. stylirostris and P. vannamei via natural routes such as cohabitation and cannibalism.
Rapid and Simple Detection of Enterocytozoon Bieneusi Using Lateral Flow Assay Based on Recombinase Polymerase Amplification or Nested PCR Combined with CRISPR-Cas12a.
Enterocytozoon bieneusi is an obligate intracellular microsporidian pathogen. It primarily causes diarrhea and weight loss in infected humans and animals, resulting in substantial economic losses to the livestock industry. Therefore, establishing a highly sensitive and specific detection method for E. bieneusi is critical for its prevention and control. crRNA and recombinase polymerase amplification (RPA) primers were designed based on partial sequences of the 18 S ribosomal RNA gene and the internal transcribed spacer (ITS) of E. bieneusi. DNA extracted from fecal samples was amplified using RPA or nested polymerase chain reaction (PCR). PCR amplicons were treated with a Tris-saturated phenol-chloroform-isoamyl alcohol mixture to obtain purified target DNA, which was subsequently introduced into the CRISPR-Cas12a reaction system. Post-reaction detection was performed via qPCR instrumentation, fluorescence visualization, and lateral flow strip (LFS) assays. The operational parameters for E. bieneusi detection were subsequently optimized using RPA/CRISPR-Cas12a or nested PCR/CRISPR-Cas12a platforms. The aforementioned methodology was concurrently validated using 50 clinical specimens with known E. bieneusi infection status. The limits of detection were 7.13 copies/µL for RPA/CRISPR-Cas12a and 2.35 × 10- 2 copies/µL for nested PCR/CRISPR-Cas12a. When the concentration of unamplified DNA in the CRISPR-Cas12a reaction system reached ≥ 1 × 10- 4 µg/µL, the single-stranded DNA reporter was efficiently cleaved, resulting in a detectable fluorescence signal. The nested PCR/CRISPR-Cas12a technology was used to analyze 50 fecal samples with confirmed E. bieneusi-positive or -negative status. The results obtained from instrument-based detection, fluorescence observation, and lateral flow test strip detection were completely consistent. We established the first integration of nested PCR with CRISPR-Cas12a for the detection of E. bieneusi. and were also the first to quantitatively explore the detection limit of Cas12a using non-amplified E. bieneusi DNA. This approach offers a rapid, specific, and highly sensitive diagnostic method. Furthermore, the wide selection of appropriate visualization methods facilitates adaptation to various laboratory conditions and sample template concentrations, enabling accurate result interpretation.
