Quebec platelet disorder

An autosomal dominant bleeding disorder due to a gain-of-function defect in fibrinolysis. Although affected individuals do not exhibit systemic fibrinolysis, they show delayed onset bleeding after challenge, such as surgery. The hallmark of the disorder is markedly increased PLAU levels within platelets, which causes intraplatelet plasmin generation and secondary degradation of alpha-granule proteins.

Gray platelet syndrome

A rare platelet disorder characterized by a selective deficiency in the number and contents of platelet alpha-granules. It is associated with mild to moderate bleeding tendency and moderate thrombocytopenia. The platelets are enlarged and have a gray appearance on light microscopy of Wright-stained peripheral blood smears due to decreased granules.

A highly malignant, metastatic, primitive small round cell tumor of bone and soft tissue that affects children and adolescents. It belongs to the Ewing sarcoma family of tumors, a group of morphologically heterogeneous neoplasms that share the same cytogenetic features. They are considered neural tumors derived from cells of the neural crest. Ewing sarcoma represents the less differentiated form of the tumors.

Hypertension-Associated Acetate Deficiency Enhances Platelet Activation and Thrombosis Via Olfr78.

Arterial thrombotic events constitute the leading cause of mortality in hypertension. Gut dysbiosis induces endothelial dysfunction and systemic inflammation, contributing to hypertension and its associated cardiovascular complications. Whether these dysbiotic microbiota metabolites in hypertension directly regulate platelet hyperactivation and thrombosis remains unclear. Fecal microbiota transplantation, 16S rRNA sequencing, and untargeted metabolomics were performed using samples from patients with hypertension. In vivo FeCl3-induced mesenteric arteriole thrombosis model, ex vivo microfluidic whole-blood perfusion assay, and in vitro platelet functional studies defined the functional effects of acetate on platelet activation. Moreover, platelet-specific Olfr78 (olfactory receptor 78)-deficient mice were employed to explore the underlying mechanisms of acetate on platelet activation. Transplantation with fecal microbiota from patients with hypertension enhanced in vivo FeCl3-injured mesenteric arteriole thrombosis and ex vivo whole blood thrombus formation compared with fecal microbiota from healthy normotensive subjects. Untargeted metabolomics revealed that gut microbiota-derived acetate was decreased in patients with hypertension, and plasma acetate concentration negatively correlated with integrin αIIbβ3 activation and P-selectin exposure. Acetate demonstrated superior antiplatelet efficacy against ADP-induced aggregation, dense-granule secretion, α-granule secretion, and integrin αIIbβ3 activation than collagen or thrombin-induced platelet activation. Mechanistic studies using platelet-specific Olfr78-/- mice revealed that acetate bound to and activated Olfr78, a receptor not previously reported to be expressed in platelets, to elevate cAMP level and activate PKA, thereby increasing p-VASP and decreasing Ca2+ mobilization as well as inactivating RhoA/ROCK2/MLC (myosin light chain) signaling to inhibit platelet activation. A high-fiber diet upregulated acetate/Olfr78 signaling in platelets to suppress microvascular thrombosis and protect against myocardial injury during myocardial infarction in mice. Acetate is a negative regulator of platelet hyperreactivity and thrombus formation via the Olfr78 receptor, and acetate deficiency contributes to platelet hyperreactivity in hypertension. Lifestyle modifications, particularly high-fiber dietary intervention and acetate supplementation, exhibit potent antithrombotic effects in hypertension.

Systems-level molecular and immunological evidence identifies Th17/Treg modulation as a key mechanism of CRSJ's neuroprotection in Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder in which neuroinflammation plays a central role. Congrong Shujing Granules (CRSJ), a traditional Chinese medicine formula, have shown clinical benefits in PD, yet their immunomodulatory mechanisms remain unclear. We investigated the effects of CRSJ on Th17/Treg immune balance. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) was used to identify representative chemical constituents of CRSJ. Representative CRSJ compounds were characterized, and their binding affinities were evaluated by molecular docking and molecular dynamics simulations. An MPTP-induced PD mouse model was established and treated with CRSJ. Behavioral outcomes, dopaminergic neuroprotection, immune cell subsets, transcriptomic profiles, and cytokine networks were assessed using flow cytometry, RNA sequencing, multiplex assays, immunofluorescence, and Western blotting. HPLC analysis identified 44 representative compounds in CRSJ spanning multiple chemical classes associated with immunomodulatory, neuroprotective, and antioxidant activities. Molecular-level prioritization of CRSJ-derived serum constituents highlighted paeoniflorin as a key Th17/Treg balance immunoregulatory candidate, exhibiting stable interactions with RORγt, Foxp3, and α-synuclein in molecular docking and molecular dynamics simulations. In an MPTP-induced Parkinson's disease mouse model, CRSJ treatment dose-dependently improved motor performance, preserved dopaminergic neurons, and reduced striatal α-synuclein accumulation. Transcriptomic profiling revealed CRSJ-associated shifts toward regulatory immune programs, characterized by attenuation of Th17-related signatures and enhancement of Treg-associated pathways, accompanied by consistent modulation of the TGF-β/SMAD3 signaling axis. These molecular changes were supported by protein-level validation. CRSJ further alleviated neuroinflammation by promoting microglial M1/M2 polarization and partially normalizing dysregulated cytokine and chemokine profiles. Integrated immunological analyses demonstrated restoration of Th17/Treg balance and suppression of CX3CL1/CX3CR1-Th17 signaling, collectively supporting an immuno-neuroprotective profile of CRSJ in PD. CRSJ exerts neuroprotective effects in PD by restoring Th17/Treg homeostasis and suppressing neuroinflammatory pathways, supporting its potential as an immunomodulatory therapy.

A lineage-specific selective autophagy receptor module mediates P-body turnover.

Processing bodies (P-bodies) are conserved ribonucleoprotein granules central to RNA metabolism across eukaryotes. Although the mechanisms underlying their assembly are well understood, the pathways governing their selective turnover remain unclear. Here, we identify the conserved decapping proteins Enhancer of mRNA decapping 4 (EDC4) and decapping protein 1 (DCP1) as a selective autophagy receptor pair responsible for P-body turnover in the model plant Marchantia polymorpha. MpEDC4 engages ATG8 via a canonical ATG8-interacting motif, while MpDCP1 contains a previously unrecognized reverse ATG8-interacting motif within its intrinsically disordered region. Mutations disrupting these motifs impair the autophagic degradation of P-bodies, demonstrating a cooperative receptor mechanism. Notably, this autophagic function is lineage-specific, as orthologs in Arabidopsis and humans lack ATG8-binding capacity. Strikingly, the heterologous expression of MpEDC4 in human cells promotes the degradation of α-synuclein, a protein linked to Parkinson's disease etiology. Our findings uncover an evolutionary innovation that links RNA metabolism to selective autophagy and open avenues for the cross-kingdom engineering of targeted protein degradation pathways.

Prevention of In-Stent Restenosis After PCI by Saponin Natural Products: Inhibition of Platelet Activation.

Percutaneous coronary intervention (PCI) enables coronary revascularisation and restores haemodynamic stability but also carries risks of delayed complications including in-stent restenosis (ISR), a chronic progressive disease with endovascular damage after PCI, which compromises the long-term efficacy of PCI. Currently, the main treatment strategies for ISR include interventional and drug therapies. From the era of using aspirin as a single antiplatelet agent to the 'gold standard' era of dual antiplatelet therapy, the risk of ISR after PCI has been reduced. However, long-term use of antiplatelet drugs inevitably causes a series of side effects such as gastrointestinal mucosal damage and bleeding, which have become key limiting factors in clinical treatment. Saponin natural products have been used to mitigate ISR progression by targeting platelet dysfunction. Specifically, these compounds directly bind to platelet membrane receptors, block ligand-receptor interactions, inhibit the secretion of α-granule and dense-granule contents, regulate intracellular signalling pathways and platelet metabolism, inhibit release of inflammatory mediators, and suppress platelet aggregation. This article reviews the progresses on the application of the active components in saponin natural products to inhibit platelet activation in the development of ISR after PCI, aiming to provide a superior approach for the comprehensive treatment of ISR after PCI.

Alphaviral Capsid Proteins Inhibit Stress Granule Assembly via Competitive RNA Binding With G3BP1.

Viral infection is one of the conditions that induce stress granule (SG) formation, a cellular defense mechanism that exerts antiviral effects. To counteract this host response, viruses have evolved a broad spectrum of strategies to inhibit SG formation. However, the molecular mechanisms underlying SG inhibition remain poorly understood. The nucleocapsid proteins play a critical role in virus replication and host interaction. Here, using Semliki Forest Virus (SFV) as a model, we uncover the function of the alphavirus nucleocapsid in SG inhibition. This inhibitory function depends on oligomerization mediated by an N-terminal α-helix and with a positively charged intrinsically disordered region (IDR). We show that SFV capsid directly competes with G3BP1 for RNA binding, thereby disrupting G3BP1-RNA liquid-liquid phase separation (LLPS) in vitro and SG assembly in cells. This mechanism is conserved across the alphavirus family but is not shared by the nucleocapsid of SARS-CoV-2 or other endemic viruses examined. Notably, expression of a peptide from SFV capsid is sufficient to inhibit SG formation induced by Amyotrophic Lateral Sclerosis (ALS)-associated mutations, suggesting potential therapeutic applications. Our findings reveal mechanistic insight into SG modulation by the viral capsid protein and provide a possible bioengineering tool for probing SG dynamics in health and disease.
