Phage P100 resistance in clinical and foodborne Listeria monocytogenes isolates is associated with adsorption-inhibiting mutations and fitness trade-offs.
Listeria monocytogenes is a common bacterial pathogen causing listeriosis. Phage-based products are increasingly used in food safety, but their use raises concerns about the emergence of phage-resistant bacteria. The broad-host-range phage P100 is applied in foods, however, little is known about the genetic basis of resistance and associated fitness trade-offs in L. monocytogenes. In this study, recently (2014-2020) circulating, clinical and food-associated L. monocytogenes strains (n = 14) were exposed to a commercial P100 phage preparation in soft agar at 20 °C and 10 °C. Eight mutants were isolated and confirmed as phage-resistant by spot assays and flow cytometry. Whole-genome sequencing revealed that most mutants derived from serovar 4b strains carried mutations in genes for polysaccharide synthesis indirectly involved in cell wall teichoic acid glycosylation, whereas mutants from serovar 2a or 2b strains had mutations in a gene directly involved in WTA glycosylation. Confocal microscopy and flow cytometry confirmed reduced phage adsorption in all mutants. The resistant mutants displayed increased antibiotic sensitivity, with two showing pronounced sensitivity to β-lactam antibiotics. These two mutants also showed impaired growth at 37 °C. Together with four additional mutants, growth was also impaired at 10 °C, both without and with sodium chloride and sodium nitrite. Overall, serovar 4b-derived mutants exhibited stronger fitness trade-offs than those of serovar 2a or 2b The findings of this study contribute to the understanding of the P100 phage-resistance mechanisms in different serovar strains of L. monocytogenes and reveal associated fitness trade-offs that may be relevant for future antimicrobial and biocontrol strategies.
Listeria Monocytogenes: A Case Series and Clinical Insights from a Tertiary Care Hospital.
Listeria monocytogenes is a rare but serious illness, especially in the immunocompromised patient. It causes invasive infection, such as bacteremia and meningitis with insidious presentation and high mortality without treatment. A cluster of six Listeria monocytogenes bacteremia was noted within 10 months at a tertiary care referral center. Patients were 27-72 years old; four also had concomitant immunosuppressive diseases, including chronic renal disease, diabetes mellitus, and cancer. All the infections were confirmed on blood culture performed with the VITEK 2 system. The organisms were all gentamicin and ampicillin-sensitive. Five patients improved on intravenous antibiotic therapy, and one died from multiorgan failure. The series reiterates the importance of maintaining listeriosis as a significant concern in febrile patients with predisposing conditions. Early diagnosis and early treatment with appropriate antibiotics have positive outcomes.
Src-dependent tyrosine-phosphorylation of NM2A has a protective role against bacterial pore-forming toxins.
Pore-forming toxins (PFTs) are key bacterial virulence factors that disrupt host plasma membrane (PM) integrity, triggering cellular stress and initiating repair mechanisms. The cytolysin Listeriolysin O (LLO), secreted by Listeria monocytogenes, has well established roles in infection, yet the host signaling responses to LLO-induced damage remain poorly understood. Here, we identify a previously unrecognized protective pathway in which LLO triggers rapid activation of the tyrosine kinase Src, leading to phosphorylation of the non-muscle myosin II heavy chain 2A (NMHC2A) at tyrosine 158. While Src activation and NMHC2A tyrosine phosphorylation have been observed during Listeria infection, we demonstrate here that both responses are directly driven by LLO. This phosphorylation event does not alter NMHC2A motor activity in vitro but is required for cytoskeletal reorganization and efficient responses to PM damage. Using Caenorhabditis elegans, we further show that phosphorylation of the NMHC2A homolog NMY-2 at the conserved tyrosine 163 is required for survival under PFT-induced stress and heat shock, revealing an evolutionarily conserved defense mechanism. Together, our findings establish Src-mediated NMHC2A phosphorylation as a critical link between PFT-induced PM damage sensing and actomyosin remodeling, advancing our understanding of host defense against bacterial toxins.
Results onListeria monocytogenesstrains from ready-to-eat meat products and food production environments in Namibia, Sudan, and Zambia: a focus on genomic characterization of strains.
Although one of the biggest listeriosis outbreaks was reported in South Africa, due to ready-to-eat (RTE) meat product, to date very few data on L. monocytogenes in foods and food environments are available from other African countries. The aims of this study were to: document L. monocytogenes presence in RTE products and food environments from Namibia, Sudan and Zambia, provide isolates genomic characterization, and evaluate genomic similarity using data available at the Italian National Reference Laboratory for L. monocytogenes database. A total of 768 samples, including RTE meat products (n=405), environmental swabs (n=228) and frozen chicken (n=135), were collected and tested for Listeria spp. and L. monocytogenes by African partners. Listeria spp. presence was observed mainly in Zambia (38.7%; n=41/106), followed by Sudan (5.0%; n=24/480) and Namibia (3.3%; n=6/182), mostly related to RTE meat products (10.1%; n=41/405). MALDI-TOF confirmed 14 of 71 strains as L. monocytogenes (n= 12 from meat RTE; n=2 from environments) and MLST identified six CCs: CC9 (n=7), CC1 (n=3), CC3 (n=1), CC37 (n=1), CC121 (n=1) and CC31 (n=1). A complete L. monocytogenes Pathogenicity Island 3 (LIPI-3) was observed in CC1 and CC3 strains, in addition to LIPI-1, which was identified in all 14 strains. Several resistance factors were detected, including stress islands (SSI-1 and SSI-2), Tn6188_qac, cadA and bcr genes. Furthermore, three cgMLST clusters were detected: two for CC9 from Zambia, one for CC1 from Sudan, all related to RTE foods. This study confirmed L. monocytogenes presence in African RTE meat products and food environments, posing a public health concern for consumers, mainly linked to CC1 strain presence, known to be a hyper-virulent clone. Moreover, the presence of L. monocytogenes strains harbouring several resistance factors, such as the CC9 clone, could help these strains to adapt, survive and persist.
Comparative Infection Pathways of Listeria monocytogenes in Humans versus Ruminants.
Severe infection caused by Listeria monocytogenes in humans is primarily linked to the pathogen's ability to cross the intestinal barrier and disseminate within the host. Once internalised, L. monocytogenes exhibits intracellular motility that enables colonisation of secondary target organs, including the brain and placenta. This review aims to critically compare the infection pathways of L. monocytogenes in humans and ruminant hosts, highlighting the molecular mechanisms that enable bacterial survival, organ tropism, and adaptation to host-specific stressors. By focusing on these comparative pathways, we aim to clarify how differences in virulence and stress-response factors, influenced by clonal complex, strain type, and lineage, affect host susceptibility and disease outcome. Using a comparative approach, we explore how variation in virulence and stress-response determinants, influenced by lineage, clonal complex, and strain type, may shape host susceptibility and disease outcomes across species. While ruminant listeriosis is well recognized clinically, key aspects of infection dynamics, tissue tropism, and host-pathogen interactions in these hosts remain poorly understood. Addressing these knowledge gaps will strengthen comparative pathogenesis frameworks and enhance the interpretation of strain diversity and host association patterns, contributing to a more integrated understanding of L. monocytogenes biology in human and animal hosts.
