Live-cell 3D-SIM of Rift Valley fever virus NSs filaments reveals a polygon web architecture.

A defining feature of Rift Valley fever virus (RVFV) is the incorporation of the NSs protein into large filamentous assemblies inside infected nuclei [R. Swanepoel, N. K. Blackburn, J. Gen. Virol. 34, 557-561 (1977).], as judged from fixed specimens. To gain insight into the 3D structure of NSs filaments within live-cell nuclei, we used genetic-code expansion (GCE) to incorporate trans-cyclooct-2-en-L-lysine into the protein. This enabled site-specific fluorescent labeling with tetrazine dyes for live-cell structured illumination microscopy (SIM). Our superresolved images revealed the complete native architecture of NSs filaments as a micron-scale polygon web of fibers with discrete domain characteristics, overturning previous assumptions of simple linear filaments. Parallel experiments on fixed RVFV-infected cells confirmed that native NSs filaments also display this morphology. Overall, our 3D-SIM analysis reveals distinct structural plasticity within NSs filaments, establishing a quantitative structure-function relationship that support the importance of polygon organization for NSs filament function during RVFV infection.

Dose-dependent immune modulation in arboviral coinfections among febrile patients.

Arboviruses pose substantial diagnostic and public health challenges in tropical regions where multiple pathogens co-circulate. Nigeria, with diverse ecological zones and limited surveillance infrastructure, represents a critical nexus for arboviral transmission, yet comprehensive data on seroprevalence, coinfection dynamics, immune modulation, and viral genetic diversity remain scarce. We conducted cross-sectional surveillance across three Nigerian ecological zones from January 2019 to December 2023. Sera from 871 participants were tested for IgG and IgM antibodies to dengue virus (DENV), chikungunya virus (CHIKV), Zika virus (ZIKV), yellow fever virus (YFV), West Nile virus (WNV), and Rift Valley fever virus (RVFV) using immunoblot assays. A stratified subset (n=300) underwent cytokine profiling for interleukin-6 (IL-6), tumour necrosis factor-alpha (TNF-α), interferon-gamma (IFN-γ), and interleukin-10 (IL-10). IgM-positive samples underwent RT-PCR confirmation with genotype assignment through sequence comparison against GenBank reference strains. Overall IgM seropositivity indicating recent infection was 86.8% (756/871), with DENV most prevalent (59.6%; 95% CI: 56.3-62.8%), followed by CHIKV (49.4%), YFV (48.3%), ZIKV (19.7%), WNV (3.4%), and RVFV (2.3%). Coinfection with two or more arboviruses was detected in 61.2% (533/871) of participants. Seropositivity was highest in the tropical rainforest zone (94.2%) compared with Guinea savanna (84.5%) and Sudan savanna (80.4%; χ²=31.8, p<0.001). Cytokine profiling revealed significantly elevated concentrations of IL-6, TNF-α, IFN-γ, and IL-10 in dual and triple infections compared with mono-infections (all p<0.001), demonstrating dose-dependent immune modulation. All four DENV serotypes were identified, with DENV-2 predominating (59.9%). Molecular characterisation confirmed circulation of CHIKV East/Central/South African genotype (82.1%), DENV-2 Cosmopolitan genotype, and both African (71.2%) and Asian (28.8%) ZIKV lineages, with study sequences showing >99% nucleotide identity to established reference strains. These findings reveal extraordinarily high levels of arboviral infection and coinfection across Nigerian ecological zones. The dose-dependent cytokine elevation in coinfections suggests distinct immunopathological mechanisms. These data underscore the urgent need for multiplex diagnostics, genotype-specific monitoring, and climate-informed vector control strategies.

Understanding the Rift Valley fever exposure risk: A comparative perspective from a multi-country study in East and Central Africa, 2021-24.

Rift Valley fever (RVF) is a concern in East and Central Africa, particularly following periods of heavy rainfall and flooding. However no human outbreaks have been reported in the Democratic Republic of the Congo (DRC). To assess whether this reflects a true absence of virus circulation, we estimated RVF seroprevalence in Goma (eastern DRC) and examined context-specific risk factors, comparing the findings with data from outbreak-prone countries. A two-year longitudinal study, across six health facilities in DRC, Kenya and Uganda, enrolled febrile subjects aged ≥10 years. Human serum samples were analyzed for RVF virus and anti-RVF antibodies. Demographic, behavioral, occupational and environmental factors were evaluated. 4,806 participants were enrolled: 1,370 (28.5%) DRC, 1,468 (30.6%) Kenya and 1,968 (40.9%) Uganda. 253 participants (5.3%) tested positive for RVF by serological and/or molecular assays: 19 (1.4%) DRC, 29 (2.0%) Kenya and 205 (10.4%) Uganda (p < 0.001). Only in Uganda, subjects tested positive for RVF virus by PCR (10 subjects, 0.5%). Occupations and activities involving contact with livestock, predominated in Kenya and Uganda, whereas handling of raw meat was most common in DRC. No specific occupations or activities were significantly associated with RVF exposure in DRC while several significant factors were identified for Kenya and Uganda. Multivariate analysis across all three countries showed that being from Uganda, male, over 20 years of age, employed as butcher or crop farmer and engaging in animal-related activities, were independently associated with RVF positivity, as was contact with sheep. Despite a prevailing sense that RVF transmission does not occur in DRC, we found a seroprevalence of 1.4%, comparable to Kenya where RVF is well documented. Further research targeting high-risk human and animal populations in DRC is warranted. A One Health approach will contribute to defining the ecology of local transmission of RVF in DRC.

Biosensors for the detection of arthropod-borne veterinary viruses: A comprehensive review.

Veterinary arboviruses pose serious threats to animal health, especially in endemic areas. Traditional methods used for the diagnosis of these viruses are expensive, time-consuming, and require specialized personnel. This review discusses biosensor technologies for veterinary arboviruses, analyzing specific targets, mechanisms, sensitivity, specificity, and utility for use in the field. The remaining gaps were also mentioned. A comprehensive literature search was carried out in the major databases from 2015 to 2025. We used the keywords "Biosensing Technologies"Arboviruses," "Veterinary Medicine," "Viral Diseases," and "Point-of-Care Systems," and names of veterinary arbovirus families and their viruses. The findings reveal that biosensors have shown great sensitivity, specificity, and fast detection for various veterinary arbovirus families in the field. The technologies used were electrochemical, optical, CRISPR-based, and microfluidic formats, which are often improved by the use of nanomaterials and smartphone integration. The platforms directed against Japanese encephalitis, Rift Valley fever, and African swine fever had the lowest detection limits of femtomolar and also had the capability for multiplexing. Nevertheless, the technological development of biosensors remains restricted to a few viruses only, thus revealing substantial veterinary diagnostic gaps. Overall, biosensors have great potential in veterinary virology, with the capability of early diagnosis and, consequently, better outbreak response. Many of these sensors are specific, sensitive, and portable, making them great options for point-of-care applications. Future attention should be given to less-studied viruses, large-scale field validation, and multitarget biosensing platforms to improve veterinary surveillance globally.

Spatiotemporal patterns of Rift Valley fever virus in Africa: a retrospective genomic epidemiology and phylodynamic modelling study.

Rift Valley fever virus (RVFV) is a mosquito-borne zoonotic pathogen causing outbreaks in humans and ruminants across Africa and the Arabian Peninsula. Originally restricted to the Great Rift Valley, RVFV has expanded geographically, prompting its classification by WHO as a pathogen of pandemic potential. We investigated the evolutionary and spatial dynamics of RVFV across Africa. We used genomic data generated at the International Livestock Research Institute Nairobi genomic laboratory (BioProject PRJNA1106221) and combined with publicly available datasets retrieved from the National Center for Biotechnology (NCBI) GenBank nucleotide database. In retrieving RVFV genome sequences from the NCBI GenBank, we applied the search terms "Rift Valley fever virus segment L AND 6404[SLEN]", "Rift Valley fever virus segment M AND 3885[SLEN]", and "Rift Valley fever virus segment S AND 1520:1690[SLEN]" for L (Large), M (Medium), and S (Small) segments, respectively. For sequences without additional spatiotemporal information, we searched PubMed to extract the associated sequence metadata. We performed molecular clock analysis, phylogenetic inference, phylodynamic modelling (continuous phylogeographic reconstruction), and landscape phylogeography on the three RVFV genome segments (L, M, and S). We aimed to assess evolutionary rates, dispersal patterns, and environmental drivers. Focus was placed on lineage C, the most widely distributed variant. The global dataset used in this study consisted of large (n=236), medium (n=237), and small (n=247), which were further filtered to exclude potential reassortants and vaccine strains. Genome sequences retrieved from NCBI GenBank database comprised large (n=180), medium (n=184), and small (n=202). The genome sequences from retrospective human and livestock isolates comprised large (n=56), medium (n=53), and small (n=45) collected in Burundi (2018), Kenya (2007, 2018, 2019, 2021, and 2022), and Rwanda (2018 and 2022). Our dataset revealed that RVFV exhibited low overall genetic diversity. Lineage C, however, showed evidence of active evolution, with substitution rates ranging from 3·58 × 10-4 to 9·76 × 10-4 substitutions per site per year. This lineage probably originated in Zimbabwe in the mid-1970s and has since expanded across eastern and southern Africa. Phylogeographic reconstructions revealed rapid spread, with diffusion coefficients exceeding 50 000 km2 per year. Lineage C appears capable of establishing endemic transmission in new regions, with ongoing diversification observed during interepidemic periods. These observations reinforce the value of continuous genomic surveillance, particularly during cryptic transmission phases when adaptive mutations might emerge. Although further evidence is needed, observed trends in climate variability and land-use change point to the potential benefit of targeted surveillance in settings that could be at increased risk, including urban centres and wetlands.
