An Fc-silent OspA monoclonal antibody passively protects mice from tick and intradermal Borrelia burgdorferi challenge.
The monoclonal antibody, LA-2, has played a pivotal role in the development of Outer surface protein A (OspA)-based vaccines for Lyme disease, a multisystem illness caused by the tick-borne spirochete, Borrelia burgdorferi sensu lato. Of particular significance was the demonstration more than three decades ago that LA-2 equivalent antibody titers, defined by a competitive-inhibition ELISA, serve as a reliable correlate of vaccine-induced protection across different species, including humans. In vitro characterization of LA-2 has identified both complement-dependent and -independent activities, although which of these attributes contribute to protection against B. burgdorferi remains unresolved. To address this issue, we generated and characterized an "Fc-silent" version of LA-2 IgG1 carrying so-called LALAPG substitutions (L234A, L235A, P329G). We demonstrate that LA-2 LALAPG retained OspA binding activity but was severely attenuated in in vitro complement deposition and complement-dependent borreliacidal assays. Nonetheless, LA-2 LALAPG was as effective as LA-2 at passively protecting C3H mice against nymphal tick-mediated B. burgdorferi sensu stricto (s.s.) B31 challenge. LA-2 LALAPG was also equivalent to LA-2 in passively protecting BALB/c mice against intradermal B. burgdorferi s.s. B31 challenge. In the intradermal challenge model, viable spirochetes were not recoverable 24 h after injection from skin biopsies of mice treated with LA-2 or LA-2 LALAPG, and an influx of pro-inflammatory cytokines and chemokines to the injection site was abrogated. Collectively, these results suggest that LA-2's primary mode of action involves direct physical interactions with the spirochete rather than complement-dependent killing. Elucidating these mechanisms may have implications for understanding the mechanistic correlates of OspA-based vaccine-induced immunity in humans.
Feasibility of an adjunctive INtervention for Debilitating symptom complexes attributed to ticks (FIND): study protocol for a randomised, waitlist-controlled feasibility trial.
Debilitating Symptom Complexes Attributed to Ticks (DSCATT) is a new term for an unexplained Australian syndrome-people who suffer from a chronic, multifaceted and debilitating illness, characteristically attributed to tick bites, but in a country without endemic Lyme disease. Despite the profound morbidity of DSCATT, no single causative agent has been identified and there are no recognised treatments for the illness at present. An increasing body of evidence shows psychological therapies such as Acceptance and Commitment Therapy (ACT) can be effective in reducing symptom-related disability and improving quality of life for other unexplained syndromes. Here we present a study protocol to assess the feasibility of an ACT-informed intervention for patients suffering from DSCATT, to be used adjunctively to their pre-existing healthcare. The study aims to assess the acceptability, practicality and demand for the treatment. Additionally, we will examine the effects of therapy on participants' health and well-being, its safety, potential mediators of response to therapy and its preliminary cost-effectiveness. We will assess the feasibility of a 32-week, randomised, waitlist-controlled, parallel convergent mixed-methods pilot trial for DSCATT. Participants will be randomised in a 1:1 ratio to receive either 16 sessions of ACT-informed therapy adjunctive to their pre-existing healthcare, delivered one-to-one with a trial therapist within a 20-week period or be assigned to the waitlist control group where they will continue their treatments as usual. We will collect quantitative and qualitative data to address study aims, with retention rate being the primary feasibility outcome. The study has ethical approval from Austin Health Human Research Ethics Committee (HREC). The outcomes will be published in peer-reviewed journals. Data from participants who have given extended consent will be available for other HREC-approved studies. Non-invasive ovulation tracking enables genetic engineering in wild rodents.
Many non-model rodent species are inaccessible to genetic engineering due to our limited understanding of their reproductive biology. Here, we present a low-cost, camera-based estrous-tracking technology that enables transgenesis in the white-footed mouse Peromyscus leucopus, a key reservoir for Lyme disease. We demonstrate the efficient generation of pregnant and pseudopregnant mice via timed ovulation, provide protocols for embryo generation, cultivation, microinjection, and transplantation as well as an accurate developmental timeline, and report the first engineered Peromyscus. The same technology successfully tracked conserved estrous-linked cycling behavior in other rodents, including hamsters. Finally, estrous tracking differentiated reproductively healthy, geriatric female Peromyscus from those with declining fertility based solely on their activity, providing a non-invasive method for studying reproductive senescence. Collectively, these tools represent a critical resource for engineering non-model rodents, advance the long-lived Peromyscus as a model organism, and will prove essential to heritably immunizing wild rodent populations against Lyme disease.
Exploring Cerebrovascular Manifestations of Neuroborreliosis: A Scoping Review.
Lyme disease is zoonotic infection transmitted by Borrelia burgdorferi and, if left undiagnosed, can present in later stages with neurological manifestations. This review aims to consolidate evidence on stroke-like or similar cerebrovascular outcomes of Lyme neuroborreliosis (LNB) to aid early recognition and treatment. A comprehensive search resulted in 90 patients meeting European Federation of Neurological Societies/Infectious Diseases Society of America/ American Academy of Neurology/American College of Rheumatology (EFNS/IDSA/AAN/ACR) criteria for LNB with radiologically or serologically confirmed cerebrovascular involvement. Data were charted descriptively. Median age was 12 years among children and 52 years in adult cases. Tick exposure was reported in 89% of cases. Presentations included hemiparesis (51%), cranial-nerve palsy (38%; Lower Motor Neuron (LMN) facial 31%), and cognitive changes (26%). Common imaging findings were vasculitis (72%) and ischaemic stroke (51%), mainly in the cortical (56%) area. Cerebrospinal fluid (CSF) lymphocytic pleocytosis (median 73 cells/µL) with presence of intrathecal Borrelia Immunoglobulin G (IgG) appeared in the majority of cases. Antibiotics (mainly ceftriaxone) were used in 85% of cases and interventional management in 13% of cases. Ninety percent of cases improved after 6 months of treatment. Mortality and persistent tetraplegia were reported in 5% and 3% of cases, respectively. LNB should be suspected in cases of cryptogenic stroke from endemic areas. Reduction in CSF pleocytosis with intrathecal Borrelia antibodies could predict a favorable antibiotic response, although some refractory cases were observed to improve only with interventional management. Prompt diagnosis and management can affect morbidity and mortality.
Telomere-to-telomere assembly detects genomic diversity in Canadian strains of Borrelia burgdorferi.
Borrelia burgdorferi, the bacteria causing Lyme disease, has a complex genome comprising a linear chromosome and multiple linear and circular plasmids. The atypical hairpin telomeres and the highly paralogous plasmids complicate genome assembly. We develop a genome assembly pipeline using both long and short reads to overcome these challenges. Using long reads, we assemble the hairpin telomeres of the linear replicons, an lp28-1a plasmid subtype, and circular plasmids of nine B. burgdorferi strains from five regions across Northwest Ontario and Manitoba, Canada. Although similar across the core conserved genomic regions, all strains carry a ∼2-10 kb right telomeric end identical to lp28-1, leading to variability in telomere length and gene content. Additionally, we observe diversity at the linear chromosome hairpin telomeric sequences, ospC types, and plasmid profiles, highlighting the genomic diversity among the geographically proximate strains and suggesting such variations as possible mechanisms of rapid evolution.
