Coal dust complexes host cell iron to impact metal homeostasis and pneumoconiosis.
Surface complexation of cell iron following particle exposure can be relevant to coal mine dust lung disease. We tested the postulate that 1) coal dust and humic substances (HS), a specific component of coal, complex intracellular iron from cultured cells to initiate a functional metal deficiency, 2) the functional cell iron deficiency which results after exposure to coal dust and HS impacts an increased release of both superoxide-related products and pro-inflammatory mediators, and 3) the disruption in iron homeostasis after coal dust exposure is associated with pneumoconiosis in miners. Cell exposures to coal dust and HS initiated a functional iron deficiency, reflected by elevated expression of an importer (divalent metal transporter-1), which increased metal uptake measured as cell non-heme concentrations. Cell exposure to coal dust and HS increased 1) generation of superoxide, measured using Nitroblue Tetrazolium reduction and an Amplex Red assay, and 2) release of interleukin (IL)-6 and IL-8. These measures of oxidative stress and inflammatory mediator release were diminished with co-exposure to iron supporting a relationship of both with a functional cell deficiency of the metal. Using a cohort of retired miners, blood ferritin levels were elevated in those diagnosed with a positive B read for coal workers' pneumoconiosis. Elevated blood ferritin concentrations correlated with progression of disease on B reads obtained one year later in the miners. It is concluded that coal dust and HS can initiate a disruption of iron homeostasis associated with superoxide generation, release of pro-inflammatory cytokines, and pneumoconiosis.
Enhanced dust removal via the synergy of a standing wave acoustic field and high-pressure spray: An integrated experimental and numerical study.
Occupational exposure to respirable coal dust poses severe health risks in underground mining operations, primarily through the development of coal workers' pneumoconiosis (CWP)-a progressive and irreversible pulmonary disease. To address this challenge, we developed an innovative dust suppression system that integrates ultrasonic atomization with acoustic agglomeration technology. The system operates via a dual-phase mechanism: ultrasonic atomization generates ultrafine water droplets (<10 μm) to form a heterogeneous dust-droplet dispersion, while high-frequency standing wave fields (20 kHz) concentrate airborne particles spatially, thereby enhancing interphase collisions between droplets and dust. The resulting agglomerates are subsequently removed by a high-pressure spray. System performance was systematically evaluated through scaled laboratory experiments that examined three critical operational parameters: nozzle orifice diameter (0.4-0.8 mm) of the high-pressure spray system, acoustic power density (60-180 W) utilized to generate the standing wave field, and duct airflow velocity (0.25-0.75 m/s). Numerical simulations integrating acoustic dynamics with CFD-DEM modeling were employed to elucidate particle trajectories and the spatiotemporal evolution of dust-droplet agglomerates within the coupled acoustic-flow field. Experimental results demonstrate a greater than 10% improvement in PM2.5 removal efficiency compared to conventional high-pressure spray systems. This enhancement is attributed to the synergistic effects of acoustic focusing and droplet entrapment. The study establishes a foundational framework for the development of acoustically enhanced pretreatment systems and offers a practical strategy for reducing respirable dust exposure in underground mining environments.
Burden of chronic respiratory disease in Asia, 1990-2023: a systematic analysis for the Global Burden of Disease Study 2023.
Chronic respiratory diseases are an important global issue, particularly in Asia, where burden patterns vary widely across countries. With more than half the world's population living in Asia, understanding the national and regional burden of chronic respiratory diseases is essential; however, research on this area remains inadequate. We aimed to investigate the burden of chronic respiratory diseases in Asia at national and regional levels, and to identify key risk factors. The Global Burden of Diseases, Injuries, and Risk Factors Study 2023 provides estimates for assessing the burden of chronic respiratory diseases, including chronic obstructive pulmonary disease (COPD), asthma, pneumoconiosis, interstitial lung disease (ILD), and pulmonary sarcoidosis. We focused on 34 countries in Asia, encompassing the high-income Asia Pacific region and central, east, south, and southeast Asia. Estimates for age-standardised prevalence and disability-adjusted life-year (DALY) rates per 100 000 population, including 95% uncertainty intervals (UIs), were extracted by location, sex, year, and Socio-demographic Index (SDI). The average annual percentage change was calculated and presented as a percentage with 95% CIs. Estimates of modifiable attributable risk factors for DALYs and mortality were also included. In Asia, the age-standardised prevalence and DALY rates for chronic respiratory diseases generally declined from 1990 to 2023; however, the trend varied substantially by disease and country. In 2023, the age-standardised prevalence rate of COPD was highest in south Asia (3044·18 [95% UI 2748·67-3303·04] per 100 000 population), while the age-standardised asthma prevalence rate was highest in the high-income Asia Pacific region (4870·24 [4046·70-5962·78] per 100 000 population) and southeast Asia (4778·18 [3970·25-5735·61] per 100 000 population). Despite southeast Asia and the high-income Asia Pacific region having a similar age-standardised asthma prevalence rate, southeast Asia had a higher age-standardised DALY rate (508·67 [95% UI 394·89-669·92] per 100 000 population) compared with the high-income Asia Pacific region (204·40 [129·23-290·41] per 100 000 population). A decrease in the age-standardised DALY rate for chronic respiratory diseases was observed with increasing SDI, contrasting with its prevalence patterns. Age-standardised DALY rates of COPD decreased in all Asian countries except for Georgia (average annual percentage change 1·37 [95% CI 1·26-1·48]) and Kazakhstan (0·73 [0·55-0·93]), and age-standardised DALY rates of asthma decreased in all countries. Smoking and ambient particulate matter pollution were identified as leading attributable risk factors for chronic respiratory diseases across Asia. Household air pollution from solid fuels was a regionally pronounced risk factor for chronic respiratory diseases, particularly in south Asia (age-standardised DALY rate 657·58 [95% UI 485·04-880·45] per 100 000 population). Although smoking was a major risk factor in males, ambient particulate matter pollution and secondhand smoke emerged as important attributable risk factors for chronic respiratory diseases in females. Countries with lower SDI had markedly higher DALY rates, highlighting the need to address socioeconomic and health-care inequities. Household air pollution from solid fuels continues to impose a substantial but preventable burden in south Asia, calling for clean energy adoption and improved ventilation. Gates Foundation.
Macrophage-Derived Ferritin Exacerbates Silica-Induced Pulmonary Fibrosis via PIK3R2-Mediated Fibroblast Differentiation.
Silicosis is a progressive and life-threatening fibrotic lung disease caused by crystalline silica. However, targeted therapies remain unavailable due to its incompletely understood pathogenic mechanisms. Here, we identify ferritin as a pivotal mediator of silica-induced pulmonary fibrosis by integrating clinical exploration with experimental validation. We detected persistently elevated ferritin levels in lung tissues and serum from silicosis patients and silica-exposed mice, and demonstrated that exogenous ferritin administration exacerbates fibrosis in vivo. Multi-omics profiling and co-culture experiments revealed that macrophage-secreted ferritin promotes fibroblast-to-myofibroblast differentiation and pathological extracellular matrix (ECM) deposition via the PIK3R2/SMAD signaling axis. Importantly, genetic knockdown of ferritin in macrophages significantly suppressed myofibroblast differentiation and collagen accumulation both in vivo and in vitro. These findings underscore that ferritin functions not only as a potential clinical biomarker for silicosis surveillance but also as a pathogenic driver through macrophage-fibroblast crosstalk, and provide a theoretical foundation for developing integrated diagnostic and therapeutic strategies against silicosis.
Toxicity of respirable coal dust: An overview of origin, chemistry, mechanisms, and possible remedies.
The prevalence of coal workers' pneumoconiosis (CWP) has been on the rise among U.S. coal miners since the early 2000s and is most pronounced in central Appalachia. Such adverse effects on human health are partly linked to the toxicity of respirable coal mine dust particles. In this review, we present an overview of the characteristics and health effects of the coal dust components, as well as the origin, chemistry, and mechanisms for their potential toxicity. Toxicity of coal mine dust is linked to the surface chemistry and bioactivity of the composing particles, such as coal, crystalline silica (quartz), pyrite, and sometimes diesel particulate matter. Formation of reactive oxygen species, such as •OH on the surface of these particles, contributes to the toxicity of coal dust. Various mechanisms including the metal-micelle coating, polymer-coating, chelation of the surface metal ions, and surface silanization, have been proposed in previous studies for the reduction or inhibition of the toxicity of coal dust particles. However, due to the complexity in surface chemistry of the various minerals and coal, there is no single universal detoxification mechanism for coal dust. One feasible remedy to address the proposed mechanisms of toxicity is the use of chemical additives as wetting agents. Important considerations for this approach include particle size and aging, mineralogy, morphology, chemical composition, solubility, surface charge, and synergistic effects of the composing phases. Additional factors to consider are the solution quality and composition, coal rank, and the potential hazards and costs associated with the chosen chemical agents.
